The simultaneous quantification of 15 mycotoxins in cow milk by liquid chromatography-mass spectrometry, is presented. Extraction was performed with acidified acetonitrile, followed by a cleanup step with sodium acetate. During validation limits of detection (LOD) and quantification, linearity, precision, accuracy, recovery, matrix effect, and stability were studied. LOD values were between 0.02 and 10.14ng/mL for aflatoxins M1, B1, B2, G1, G2, ochratoxins A and B, HT-2 and T-2 toxins, deepoxy-deoxynivalenol, zearalenone, sterigmatocystin and fumonisins B1, B2 and B3. Recovery values were between 82.6 and 94.4% for all the mycotoxins, except for fumonisins. The recovery values for fumonisins were between 42.1% and 64.6%. Matrix effect, between 25.5 and 96.8%, appeared for all of the mycotoxins, especially for deepoxy-deoxynivalenol, zearalenone and sterigmatocystin. The validated method achieves the quantification of those mycotoxins of major concern and mycotoxins that are not frequently studied in milk, such as fumonisins, sterigmatocystin or ochratoxin B.

A high-throughput LC-MS/MS method for simultaneous identification and quantitation of eight non-steroidal anti-inflammatory drugs (NSAIDs) and chloramphenicol (CAP) in bovine and ovine milk was developed. The method is capable of detecting and quantitating: Carprofen (CPF), tolfenamic acid (TLF), 5-hydroxy flunixin (FLU-OH), diclofenac (DCL), 4-methylaminoantipyrin (4MAA), meloxicam (MLX), ibuprofen (IBU), phenylbutazone (PBZ) and chloramphenicol (CAP) at their maximum residue limits (MRLs) or target analytical levels. Addition of ascorbic acid into the milk samples before the extraction step was found to be crucial for repeatability and intensity of PBZ and 4MAA. The method consists of the single extraction step with acetonitrile and a commercially available salt mixture, followed by evaporation of the supernatant and reconstitution. Complementary LC-MS/MS and LC-MS/MS/MS methods were developed for confirmation of ibuprofen and diclofenac respectively. The method was validated according to EU Commission Decision 2002/657/EC requirements. The method accuracy was in the range of 89-108% and coefficients of variation of the interday precision assessment varied between 3% and 16%. This method has been implemented at the Israel National Residue Control Laboratory for routine monitoring of NSAIDs and CAP residues in bovine and ovine milk.

Multi-class chemical contaminants, such as pesticides and mycotoxins, are recognized as the major risk factors in agro products. It is thus necessary to develop rapid and simple sensing methods to fulfill the on-site monitoring of multi-class chemical contaminants with different physicochemical properties. Herein, a lateral flow immunoassay via time-resolved fluorescence was developed for the rapid, on-site, simultaneous, and quantitative sensing aflatoxin B1 (AFB1), zearalenone (ZEA), and chlorothalonil (CTN) in maize and peanut. The sample preparation was optimized to a single step, combining the grinding and extraction. Under optimal conditions, the sensing method lowered the limits of detection (LOD) to 0.16, 0.52, and 1.21 µg/kg in maize and 0.18, 0.57, and 1.47 µg/kg in peanut with an analytical range of 0.48–20, 1.56–200, and 3.63–300 µg/kg for AFB1, ZEA and CTN, respectively. The protocol could be completed within 15 min, including sample preparation and lateral flow immunoassay. The recovery range was 83.24–110.80%. An excellent correlation was observed between this approach and high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) for mycotoxins and gas chromatography-tandem mass spectrometry (GC-MS/MS) for pesticide in maize and peanut. This work could be applied in on-site multi-class sensing for food safety.
