Resultados. El 9,1 % de los perros de la muestra fueron seronegativos y el 25,1 % no evidenciaron respuesta inmunitaria humoral adecuada a la vacunación. La concentración de anticuerpos disminuyó gradualmente desde la aplicación de la vacuna, y estuvo asociada a edad y calidad de las vacunas, aunque no estuvo asociada a sexo ni a raza. Conclusiones. Con el fin de aumentar los porcentajes de perros seropositivos y con respuesta inmunitaria humoral adecuada, se hacen las siguientes recomendaciones: 1) utilizar vacunas antirrábicas viables; 2) aplicar dos dosis de vacuna durante los primeros seis meses de vida de los cachorros; 3) aplicar refuerzos de vacuna, por lo menos, una vez al año; 4) que las autoridades vigilen las actividades y los procesos programáticos relacionados con la vacunación antirrábica por particulares.Palabras clave: vacunas antirrábicas, virus de la rabia, Lyssavirus, zoonosis, vigilancia epidemiológica, inmunidad, Colombia. Evaluation of the seroconversion as a response to rabies vaccination in dogs, Valle del Cauca, Colombia, 2009Introduction. The province of Valle del Cauca has been free of dog rabies for more than 20 years. However, sylvatic rabies foci remain which are threats to the health of the populace and its pets. Rabies vaccination campaigns are carried out annually in all 42 counties of the province. Objectives. The impact of dog vaccination was evaluated on the basis of humoral immunoresponse, population parameters and correlation with variables inherent to the vaccination process and logistics. Materials and methods. Sera and associated data were obtained from each of the 42 counties for a total sample of 569 rabies-vaccinated dogs. Rabies neutralizing antibodies were measured by quantitative ELISA. The data were analyzed with the statistical programs in Epi-Info 6.0.Results. Nearly 10% of dogs were seronegative (9.1%) and an additional 25.1% did not elicit an adequate humoral immune response to vaccination. Concentration of rabies neutralizing antibodies diminished gradually with the time after vaccination and was correlated with dog age and vaccine quality. No associations were noted between dog gender or breed. Conclusions. These data permit the following recommendations: (1) only viable, non expired rabies vaccines must be used to immunize animals, (2) two doses of rabies vaccine must be applied during the first six months of dog life, (3) booster immunizations must be administered every year, (4) practices

Background: Rabies virus infection causes encephalitis, which is almost always fatal. Vaccination can be extremely effective at preventing disease but is prohibitively costly. Vaccine formulations allowing dose-sparing and fewer inoculations with faster antibody response would be extremely desirable. IMT504, an immunostimulatory non-CpG oligodeoxynucleotide, is a highly potent vaccine adjuvant. Methods: Human and rat antibody measurements, and rat challenge studies were performed. Results: In rats, highly effective immune responses with IMT504 were observed even after diluting vaccine up to 1/625. In highly lethal, live intracerebral rabies challenge studies, protection occurred even with extremely dilute vaccine plus IMT504. In humans, antibody titers developed faster and were significantly higher with IMT504-adjuvanted diluted vaccine vs non-adjuvanted vaccine (full strength or diluted). All five administered IMT504-adjuvanted diluted vaccine reached protective antibodies (≥0.5 IU/ml) after the second injection. After the third injection, individuals receiving IMT504-adjuvanted diluted vaccine reached levels approximately 10 times higher than controls (M ± SEM: 31.0 ± 10.9 vs 3.40 ± 0.99 IU/ml). Conclusions: These data suggest that IMT504 may allow fewer inoculations, highly significant dose-sparing of vaccine, rapid antibody production and protection from rabies. Extensive clinical studies are necessary to confirm if the use of IMT504 will permit significantly greater access to highly effective life-saving rabies vaccines.

The molecular mechanisms associated with rabies virus (RV) virulence are not fully understood. In this study, the RV Flury low-egg-passage (LEP) and high-egg-passage (HEP) strains were used as models to explore the attenuation mechanism of RV. The results of our studies confirmed that the R333Q mutation in the glycoprotein (G R333Q ) is crucial for the attenuation of Flury RV in mice. The R333Q mutation is stably maintained in the HEP genome background but not in the LEP genome background during replication in mouse brain tissue or cell culture. Further investigation using chimeric viruses revealed that the polymerase L gene determines the genetic stability of the G R333Q mutation during replication. Moreover, a recombinant RV containing the LEP G protein with the R333Q mutation and the HEP L gene showed significant attenuation, genetic stability, enhancement of apoptosis, and immunogenicity. These results indicate that attenuation of the RV Flury strain results from the coevolution of G and L elements and provide important information for the generation of safer and more effective modified live rabies vaccine.Rabies virus (RV) belongs to the genus Lyssavirus of the family Rhabdoviridae and causes a fatal neurological disease in humans and animals (6). The RV genome is a nonsegmented negative-strand (NNS) RNA encoding five structural proteins: nucleoprotein (N), phosphoprotein (P), matrix protein (M), glycoprotein (G), and large polymerase (L). Among these, the G protein is a major contributor to RV pathogenicity (7,31,33). The G protein facilitates fast virus entry and transsynaptic spread and regulates the rate of virus replication, together with other viral elements (8,30,39). The G protein of nonpathogenic RV strains can trigger apoptosis, while the RV G of pathogenic strains induces less or no apoptosis (35, 59). The amino acid residue at position 333 of the G protein (G333) of some fixed strains has been shown to be an important determinant of virulence in adult mice (5). Strains that have arginine or lysine at position G333 kill adult mice, whereas mutants with other amino acids at this site cause a nonlethal infection (1,5,25,36,49,53). However, the pathogenicity of RV strains is not solely determined by substitutions at the G333 position. Other substitutions in the G protein, such as N194K, have also been shown to affect viral pathogenicity in mice (10, 21, 50). In addition, other viral elements, such as the N, P, M, and L genes, the trailer sequence in the noncoding region, and the pseudogene, were also reported to modulate RV pathogenicity (12,46,57,58).
