Multicenter phase II study of weekly docetaxel, cisplatin, and S-1 (TPS) induction chemotherapy for locally advanced squamous cell cancer of the head and neck

Abstract: BackgroundThe purpose of this study was to evaluate the efficacy and tolerability of weekly docetaxel, cisplatin, and S-1 (weekly TPS) as induction chemotherapy for patients with locally advanced head and neck squamous cell carcinoma (HNSCC).MethodsA total of 35 patients with previously untreated, locally advanced HNSCC were enrolled. Seven patients (20%) were diagnosed with stage III HNSCC and 28 patients (80%) were diagnosed with stage IV. Induction treatment included 30 mg/m2 docetaxel on day 1 and 8, 60 mg… Show more

“…The 5-fluorouracil (5-FU) has been replaced by capecitabine, 29 ifosfamide, 24,26 cetuximab, 23 and gemcitabine. 34 Other authors have replaced docetaxel with paclitaxel 24,25,27,28 or split the docetaxel dose (biweekly docetaxel) 35 to reduce hematological toxicity, whereas others omit the third agent. 36 By contrast, the TPF regimen affects the administration of definitive locoregional treatment in the majority of patients with locally advanced HNSCC.…”

“…This injection schedule mirrors the clinical setting where, for example, patients receive weekly chemotherapy. 3133 Also, longitudinal imaging protocols to assess treatment success may be carried out at similar time frames. We sought to determine PVX-specific serum IgM and IgG antibody levels resulting from the repeat administrations of PVX.…”

“…The value contributed by the addition of 5‐FU into an induction chemotherapy regimen is controversial. Although most induction trials have used TPF triple therapy, a small set of articles have been published using TP‐based induction chemotherapy . These articles show similar efficacy and substantially lower toxicity profiles, therefore improving the viability and adherence to induction protocols.…”
