VETERINARY DRUG RESIDUES IN MILK AND EGGS. The use of veterinary drugs in food producing animals may result in the presence of residues in foods, including milk and eggs. Immunoassay or microbiological tests are used to screen for residues, but chromatographic methods are needed to confirm positive results. In most methods, the sample is extracted with acetonitrile, submitted to clean up or directly analyzed by LC-MS/MS. Results of the Brazilian governmental monitoring programs from 2006 to 2011 have shown that the antiparasitic ivermectin was the drug most frequently found in milk. Residues in eggs are only monitored by one of the programs, and few studies have reported the incidence of veterinary drugs in this matrix in Brazil.Keywords: veterinary drugs; milk; eggs. INTRODUÇÃOMedicamentos são usados na medicina veterinária no tratamento e prevenção de doenças ou como promotores de crescimento. 1,2 Estes compostos compreendem uma variedade de classes químicas com ações terapêuticas diversas, incluindo antibiótica/antimicrobiana, antiparasitária, inseticida, fungicida e sedativa. 3 Os antimicrobianos (b-lactâmicos, aminoglicosídeos e tetraciclinas) e os antiparasitários (avermectinas, piretróides e organofosforados) são os produtos mais comercializados no Paraná para tratamento do rebanho leiteiro. 4 O uso de medicamentos veterinários em animais produtores de alimentos pode deixar resíduos nos alimentos, como carne, leite e ovos, cujos níveis não devem ultrapassar o Limite Máximo de Resíduos (LMR). O LMR é a concentração máxima de resíduo em um alimento de origem animal resultante do uso de um medicamento veterinário (expresso em mg/kg ou µg/kg, em peso fresco) que é recomendado para ser legalmente permitido ou reconhecido como sendo aceitável no alimento. 5 Estes limites são estabelecidos para garantir o uso adequado destes produtos, limitar a exposição e proteger a saúde dos consumidores dos alimentos provenientes de animais tratados. 5 A presença de resíduos em um alimento acima do LMR estabelecido indica que o produto não foi utilizado segundo as boas práticas de uso de medicamentos veterinários (BPMV), ou o uso de um produto não autorizado. 6 O Brasil não estabelece LMR para medicamentos veterinários, adotando aqueles recomendados pelo Mercosul, Codex Alimentarius, União Européia ou Estados Unidos. 7 A exposição humana a resíduos de medicamentos veterinários presentes nos alimentos pode causar efeitos adversos, incluindo reações alérgicas em indivíduos hipersensíveis 8 e câncer. 9,10 Adicionalmente, a exposição a antimicrobianos/antibióticos pode provocar o desenvolvimento de microrganismos resistentes, que dificulta a ação terapêutica destes medicamentos em indivíduos que consumiram alimentos de animais tratados. 11,12 Resíduos de medicamentos veterinários no leite podem também causar problemas tecnológicos nos processos de fermentação dos laticínios. 13,14 No Brasil, há dois programas nacionais que monitoram a presença de resíduos de medicamentos veterinários em produtos de origem animal. A multiclass method for screening and confirmatory analysis of antimicrobial residues in milk has been developed and validated. Sixty-two antibiotics belonging to ten different drug families (amphenicols, cephalosporins, lincosamides, macrolides, penicillin, pleuromutilins, quinolones, rifamycins, sulfonamides and tetracyclines) have been included. After the addition of an aqueous solution of EDTA, the milk samples were extracted twice with acetonitrile, evaporated and dissolved in ammonium acetate. After centrifugation, 10 µl were analysed using LC-Q-Orbitrap operating in positive electrospray ionization mode. The method was validated in bovine milk in the range 2-150 µg kg(-1) for all antibiotics; for four compounds with maximum residue limits higher than 100 µg kg(-1) , the validation interval has been extended until 333 µg kg(-1) . The estimated performance characteristics were satisfactory complying with the requirements of Commission Decision 2002/657/EC. Good accuracies were obtained also taking advantage from the versatility of the hybrid mass analyser. Identification criteria were achieved verifying the mass accuracy and ion ratio of two ions, including the pseudomolecular one, where possible. Finally, the developed procedure was applied to 13 real cases of suspect milk samples (microbiological assay) confirming the presence of one or more antibiotics, although frequently, the maximum residue limits were not exceeded. The availability of rapid multiclass confirmatory methods can avoid wastes of suspect, but compliant, raw milk samples. Copyright © 2016 John Wiley& Sons, Ltd.

In the aqueous Britton-Robinson buffer (pH 5.0-9.0) that served as the supporting electrolyte, one reduction peak of EES was observed in the investigated potential range between -0.75 V and -1.80 V vs SCE, with peak potential maxima ranging from -1.59 V to -1.70 V, which strongly depended on the applied pH, as did the peak shape. For the analytical purposes the pH of 7.0 was selected, since in this electrolyte the EES peak was well-shaped and separated from the background current of the supporting electrolyte in both cases; in the direct cathodic SWV and in the case of square wave adsorptive stripping voltammetry (SW-AdSV). It was established, by the Ep-pH correlation, that protons strongly influenced the electrochemical reduction of EES. The CVs recorded between 0.025 -0.50 V s -1 at pH 7.0 confirmed that the electrode reaction is adsorption-controlled. Based on the series of 1 H NMR measurements it is proved that the tertiary amino group of EES is mainly in its protonated form at pH 7.0 which may lead, at appropriate accumulation potential and time, to the favored adsorption of the target ionic form of the analyte improving on such way the sensitivity of the SW-AdSV method.The optimized procedures resulted in stable SWV responses with good linear correlation in the EES concentration range from 4.53 to 29.8 µg mL -1 (LOD = 1.36 µg mL -1 ), and from 0.69 µg mL -1 to 2.44 µg mL -1 (LOD 0.21 µg mL -1 ) in the case of optimized SWAdSV. The relative standard deviation is below 1.5%. The reliability of the elaborated procedures and thus the accuracy of the obtained results were validated by comparing them with those obtained by means of HPLC-DAD measurements.
