Multidrug-resistance-associated protein MGr1-Ag is identical to the human 37-kDa laminin receptor precursor

Abstract: We report the isolation and functional characterization of the gene encoding MGr1-Ag, a multidrug-resistance-associated protein. A lambdagt11 cDNA library derived from colorectal carcinoma SW480 cells was screened with monoclonal antibody MGr1. DNA homology analysis of 22 positive clones (designated R1-R22) suggested human 37-kDa laminin receptor precursor (37LRP, R7/R9/R15/R16/R19/R20) and a novel gene (R22) as candidate genes encoding MGr1-Ag. Western blot analysis showed that anti-R20 serum reacted with a u… Show more

“…Drug sensitivity was evaluated using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay as described previously (16). Briefly, cells were diluted with culture medium to the seeding density (1 Â 10 4 /well), suspended in 96-well flat-bottomed plates (200 AL/well; Costar, Cambridge, MA), and incubated at 37jC for 24 hours.…”

“…23) The laminin receptor takes part in the mediation of multidrug resistance in gastric cancer cells by interferring with drug accumulation and cell apoptosis. 24) Heat-shock proteins (HSPs) are ubiquitous and highly conserved proteins whose expression is induced in response to a wide variety of physiological and environmental stresses. Several HSPs have been found to interact with various components of the programmed cell death machinery and play a role in the proteasome-mediated degradation of client proteins.…”
