Multiloci analyses suggest synonymy among Rhomboplites, Ocyurus and Lutjanus and reveal the phylogenetic position of Lutjanus alexandrei (Lutjanidae: Perciformes)

Abstract: Lutjanidae comprises 21 genera and 135 species widespread throughout Atlantic, Indian and Pacific oceans. Nonetheless, the phylogenetic relationships of Lutjaninae remain uncertain. Furthermore, phylogenetic hypotheses for Lutjanus alexandrei, an endemic species from northeastern Brazilian coast, in Lutjanidae are absent so far. Therefore, we carried out multiloci analyses, combining both mitochondrial and nuclear DNA sequences in Lutjaninae species from Western Atlantic focusing on the controversial relations… Show more

“…The molecular characterization of fish samples using COI gene sequences and the phylogeny, as revealed in this study, represents one of the most comprehensive phylogenetic coverages of Lutjanus species (33 species) compared with other phylogenetic studies on Lutjanus [47][48][49][50]; however, it could not cover every species of Lutjanus due to limited genetic information of the existing species (n = 180). The strongly supported phylogenetic positions of L. bohar and L. argentimaculatus showed the reliability of this molecular tool in the identification of CFP-related fish species.…”

“…The aligned sequences were submitted to the public platforms GenBank NCBI (National Center for Biotechnology Information— http://www.ncbi.nlm.nih.gov ) [ 45 ], using the BLAST search tool (Basic Local Alignment Search Tool), and BOLD Systems (Barcoding of Life Database— http://www.barcodinglife.org ) [ 46 ] to confirm the taxonomic identification of samples. To improve the molecular identification analysis, public sequences available for Lutjanidae species in Brazil were included [ 40 , 47 , 48 ]. Sequences of Genyatremus luteus (Haemulidae) were used as an outgroup.…”

“…Therefore, the BPP requires a specified guiding tree, as well as an input file with the nominal species as defined by the user and a multiloci dataset (Cytb, D-loop, S7-2 and Myo-2). In this study, we tested the following topology, based on the results of previous studies (Veneza et al 2019, Gold et al 2011): (((L. purpureus, L. peru), L. vivanus), L. buccanella). The BPP analysis comprised 100,000 generations with sampling at every five generations and using a burn-in of 40,000.…”

“…Most of phenotypic traits used to identify the species of red snappers are highly similar, such as dorsal and anal fin spines and rays, lateral line scales, and color pattern (Cervigón 1993, Cervigón et al 1992). In addition, the genetic studies have provided conflicting results (Chu et al 2013, Gold et al 2011, Sarver et al 1996, Veneza et al 2019. According to Gold et al (2011), the contradiction between these studies are derived from the utilization of different approaches and incorporation of plesiomorphic traits.…”
