The propagation of spontaneous action potentials in 7-9 somite embryonic pre-contractile chick hearts was measured optically using a potential-sensitive merocyanine-rhodanine dye. Spontaneous optical signals, corresponding to action potentials, were recorded simultaneously from 8-16 different sites of the primitive embryonic heart. Short delays were observed in the time of occurrence of optical signals obtained from the different regions. We have found (i) switching phenomena: the site exhibiting pacemaking priority was first situated in the right pre-atrium, thereafter it switched over to the left pre-atrium, or vice versa, and (ii) double pacemakers : two different pacemaking areas were situated independently in the right and left pre-atrial portions of the heart. On the basis of analysis of such behavior, it was concluded that the regional priority of the pacemaking activity is not rigid but is flexible, that and the direction of the spread of excitation is adaptable to the circumstances in the early embryonic heart. A B STRA C T Modulation of the amount of neuropeptide released from a neurosecretory tissue may be achieved by different means. These include alterations in the quantity secreted from each active nerve terminal or in the actual number of terminals activated. From the vertebrate hypothalamus, magnocellular neurons project their axons as bundles of fibers through the median eminence and infundibular stalk to arborize extensively and terminate in the neurohypophysis, where the neurohypophysial peptides and proteins are released into the circulation by a Ca-dependent mechanism. Elevating [Ca2+]o increases the magnitude of an intrinsic optical change in the neurohypophysial terminals that is intimately related to the quantity of neuropepfide released. Similarly, the addition of micromolar concentrations of 4-aminopyridine to the bathing solution enhances this change in large angle light scattering. However, we show here that, while these effects are superficially similar, they reflect different mechanisms of action. Evidence from intrinsic optical signals (light scattering) and extrinsic (potentiometric dye) absorption changes suggests that calcium increases the amount of neuropeptide released from each active terminal in the classical manner, while 4-aminopyridine exerts its secretagogue action by enhancing the invasion of action potentials into the magnocellular neuron's terminal arborization, increasing the actual number of terminals activated. Physiologically, electrical invasion of the complex terminal arborization in the neurohypophysis may represent an extremely sensitive control point for modulation of peptide secretion. This would be especially effective in a neurohaemal organ like the posterior pituitary, where, in contrast with a collection of presynaptic terminals, the precise location of release is less important than the quantity released.

SUMMARY1. Electrical activity in the embryonic chick brain stem has been monitored optically. The vagus-brain stem preparations isolated from 7-day-old chick embryos were stained with voltage-sensitive merocyanine-rhodanine dyes.2. Voltage-related optical absorption signals evoked by vagus nerve stimulation with depolarizing and hyperpolarizing pulses using a suction electrode were recorded simultaneously from 127 adjacent loci in the brain stem using a 12 x 12-element photodiode array.3. The optical signals evoked by the stimulation appeared to be concentrated longitudinally in the central region and in the lateral region, both on the stimulated side of the brain stem, and they did not spread to the opposite side. In addition, the evoked optical responses were detected from small areas on the dorsal surface of the stimulated side, in experiments using transverse slices of brain stem. 4. The optical action potential signals evoked by the brief depolarizing stimulus were conducted slowly and were blocked completely by tetrodotoxin. With relatively long-duration depolarizing and hyperpolarizing stimulations, electrotonic responses were recorded. 5. When 2 1sA/2 ms hyperpolarizing pulse stimulations were applied, anode-break excitation signals were detected, and these signals were also blocked by tetrodotoxin.6. On the basis of the data obtained from these experiments, we constructed maps of the electrical response area and demonstrated the spatial pattern of the vagus dorsal nucleus in the 7-day-old embryonic chick brain stem.
