Multiplex Detection of Aspergillus fumigatus Mycoviruses

Abstract: Mycoviruses are viruses that naturally infect and replicate in fungi. They are widespread in all major fungal groups including plant and animal pathogenic fungi. Several dsRNA mycoviruses have been reported in Aspergillus fumigatus. Multiplex polymerase chain reaction (PCR) amplification is a version of PCR that enables amplification of different targets simultaneously. This technique has been widely used for detection and differentiation of viruses especially plant viruses such as those which infect tobacco, … Show more

“…A. fumigatus is regularly reported to be infected by viruses (592,593). The presence of these viruses may have a role in fungal virulence, since the sequenced strain Af293, which is infected with a mycovirus, is less pathogenic than other wild-type strains (176).…”

“…Moreover, sputa collected from CF patients contained high number of phages which can be stimulated by biofilm or hypoxia conditions during bacterial growth [19]. Many A. fumigatus strains are strains host viruses intracellularly suggesting a putative role in horizontal gene transfer [20].…”

“…However, these methods could not distinguish the four P. ostreatus mycoviruses in one reaction. Multiplex RT-PCR has become a powerful tool for the simultaneous detection of several different viruses at a time [ 39 , 40 ]. In the present study, we developed a multiplex RT-PCR assay for the simultaneous detection of the four known P. ostreatus viruses.…”
