Multistep Denaturation ofBorrelia burgdorferiOspA, a Protein Containing a Single-Layer β-Sheet

Abstract: The crystal structure of a soluble form of Borrelia burg-dolferi outer surface protein A (OspA) complexed with the Fab fragment of a monoclonal antibody has revealed an unusual structure that has a repetitive antiparallel P topology with a nonglob-ular, single layer P-sheet connecting the globular N-and C-terminal domains. Earlier NMR studies have shown that the local structure of OspA including the single layer P-sheet is similar to the crystal structure. Here we report a small angle X-ray scattering (SAXS) s… Show more

“…In greater detail, SEED decomposition of (i-iii) T4 phage lysozyme, α-lactalbumin, and OspA is consistent with the experimentally determined boundaries of intermediate structures (31)(32)(33)(34). (iv) β-Lactamase was found to unfold into two equilibrium intermediates with a concurrent decrease in the helical CD signal of each (35), consistent with SEED decomposition into a threedomain protein with two small partially helical domains.…”

“…A 66-residue, amino-terminally cystine-linked coiled coil derived from the leucine zipper region of the protein GCN4 was synthesized and purified by analogy to Choma et al (19). Staphylococcal nuclease (Snase), ␣ subunit of tryptophan synthase (␣-TS), and outer surface protein A (OspA) were gifts of D. Shortle (The Johns Hopkins University, Baltimore), O. Bilsel (University of Massachusetts Medical School, Worcester), C. R. Matthews (University of Massachusetts Medical School), and S. Koide (University of Chicago, Chicago) (6,20). Ubiquitin was expressed from the wild-type human expression vector pRSUB (Entrez gi576323), which was mutated (F45W), expressed, and purified as described in ref.…”

“…The determined R g is within the range observed for other proteins of similar length. For example, the ␣-subunit tryptophan synthase is 268 residues long and has an R g of 19.1 Å, whereas OspA has an R g of 25.0 Å and is 257 residues long (44,45). A homodimer of E4-ORF3 contains 272 amino acid residues, including the unstructured residues in the His 6 tag.…”
