Multivesicular body morphogenesis requires phosphatidyl-inositol 3-kinase activity

Abstract: Multivesicular bodies are endocytic compartments containing multiple small vesicles that originate from the invagination and 'pinching off' of the limiting membrane into the luminal space [1] [2] [3]. The molecular mechanisms responsible for the formation of these compartments are unknown. In the human melanoma cell line Mel JuSo, newly synthesised major histocompatibility complex (MHC) class II molecules accumulate in multivesicular early lysosomes [4]. The phosphatidylinositol (PI) 3-kinase inhibitor wortman… Show more

“…Note: we observed that cells treated with 80 nM wortmannin exhibited vesicular-like structures that stained for both PAK1 and F-actin. These may represent the swelling of vesicular compartments previously reported to be induced by wortmannin treatment [45].…”

“…45,46 PI3K inhibition thus limits MVB maturation. [46][47][48][49] WT and PKD1 − / − 3 − / − GFP-CD63 + DT40 cells were incubated alone or with the PI3K inhibitor LY294002. Inhibitor-treated WT DT40 cells formed a cluster of enlarged, ring-shaped CD63 + vesicles similar to those found constitutively in PKD1 − / − 3 − / − DT40 cells (Figure 6a).…”

“…Morphologically, wortmannin induces the vacuolation of BP-80-labeled PVCs/MVBs in BY-2 cells (Tse et al, 2004;Lo and Jiang, 2006), and it seems that such a vacuolation is a general response to wortmannin treatment in various plant cell types and species (Miao et al, 2006;Tse et al, 2006). Wortmannin inhibits the formation of the internal vesicles in multivesicular endosomes in both mammalian and plant cells and causes these organelles to enlarge significantly (Fernandez-Borja et al, 1999;Bright et al, 2001;Tse et al, 2004). The source of the extra membrane required for this dilation has not been identified, but it has been calculated that it exceeds the amount predicted to arise from a lack of internal vesicle production (Futter et al, 2001).…”
