Murine monoclonal antibodies to Klebsiella pneumoniae protect against lethal endotoxemia and experimental infection with capsulated K. pneumoniae

Abstract: To prepare monoclonal antibodies (MAbs) directed against the core-lipid A fractions of smooth lipopolysaccharide (LPS) from Klebsiella pneumoniae O1:K2, we immunized BALB/c mice with the LPS-associated proteins plus LPS. This preparation exposed the core-lipid A moiety, which is normally hidden in the micellar structure of classical LPS preparations. Among 10 MAbs selected for their reactivity with LPS-associated proteins plus LPS from K. pneumoniae O1:K2, 6 (3A3, 3C2, 3C4, 7D2, 11C3, and 12B6) were directed a… Show more

“…During the last decade, numerous murine and human MAbs that recognize common core structures within the LPSs of gram-negative bacteria have been developed (1,5,19,33,38,44,47). Some of these MAbs have been shown to react with individual Klebsiella strains that were not characterized with respect to their 0 group (5,38,44).…”

“…A multivalent LPS vaccine composed of these eight O antigens or at least the inclusion of the common O1 antigen in a broad spectrum capsular polysaccharide vaccine might be a promising approach. Recently, the administration of monoclonal antibodies to the Klebsiella O1 antigen has been reported to be protective in a mouse model of lethal endotoxemia (136). Moreover, the inclusion of O antigens in a multivalent Klebsiella vaccine formulation might be of additional benefit due to strong adjuvant action, as has been demonstrated for the Klebsiella O3 lipopolysaccharide (259).…”

“…Klebsiellae are the second most common microorganisms isolated, after E. coli, from gram-negative bacteremia patients (11,17). Al-though the structures of the nine known 0 antigens (13,16,27) have been elucidated, very little research has been done on the core and immunochemistry of Klebsiella LPS and very few MAbs against its core and lipid A have been reported (20).…”
