Murine Oncostatin M Stimulates Mouse Synovial Fibroblasts in Vitro and Induces Inflammation and Destruction in Mouse Joints in Vivo

Abstract: Oncostatin M (OSM) is a multifunctional cytokine, a member of the interleukin-6/leukemia inhibitory factor (IL-6/LIF) family, that can regulate a number of connective-tissue cell types in vitro including cartilage and synovial tissue-derived fibroblasts, however its role in joint inflammation in vivo is not clear. We have analyzed murine OSM (muOSM) activity in vitro and in vivo in mouse joint tissue, to determine the potential role of this cytokine in local joint inflammation and pathology. The effects of muO… Show more

“…For example, local over-expression of OSM in the mouse knee joint induces changes to the joint that resemble OA, including cartilage destruction and periosteal bone formation similar to osteophytes (98,99). OSM also induces proteoglycan loss and cartilage damage ex-vivo (97,100).…”

“…A common theme in connective tissue remodeling and homeostasis is the balance between the actions of matrix metalloproteinases (MMPs) and tissue inhibitors of MMPs (TIMPs). Oncostatin M has a prominent role in the regulation of connective tissue extracellular matrix (ECM) through regulation of MMPs and TIMPs (8)(9)(10)(11)(12). Utilizing an adenoviral vector (Ad) overexpression system, we previously demonstrated that transient ectopic expression of mouse (m)OSM (AdmOSM) in the lungs of mice leads to ECM accumulation (13,14).…”

“…Intraarticular overexpression of OSM in transgenic mice causes joint inflammation and cartilage destruction (15,16). Furthermore, the relevance of OSM in arthritis was shown in a murine model in which application of antibodies to OSM ameliorated arthritis (17).…”
