Mutagenicity testing of nine herbicides and pesticides currently used in agriculture

Abstract: Nine herbicides and pesticides were tested for their mutagenicity using the Drosophila sex-linked recessive lethal mutation assay. These are Ambush, Treflan, Blazer, Roundup, 2,4-D Amine, Crossbow, Galecron, Pramitol, and Pondmaster. All of these are in wide use at present. Unlike adult feeding and injection assays, the larvae were allowed to grow in medium with the test chemical, thereby providing long and chronic exposure to the sensitive and dividing diploid cells, i.e., mitotically active spermatogonia and… Show more

“…The result of the present study with a plant assay system agreed with another study with an insect assay system where it was found that exposure to Ambush (a permethrin-containing insecticide) during larval development increased sex-linked lethal mutations in fruit flies (Kale et al, 1995).…”

“…Industrial effluents, agricultural runoff, and municipal wastewaters contain unknown substances and complex mixtures that are released into the environment and can lead to contamination of surface and subsurface waters [Houk, 1992;Kale et al, 1995;White et al, 1996]. Compounds present in polluted water are capable of causing biological alterations that can affect particular populations and entire ecosystems [Rajaguru et al, 2003].…”

“…Thus, although a first report did not obtain positive induction in the sexlinked recessive lethal test after treating adult flies [Vogel and Chandler, 1974], other authors have reported slight mutagenicity in the same test following this procedure [Magnusson et al, 1977]. On the other hand, whilst no mutagenicity was obtained when the chemical was administered by feeding and/or by injection to adults [Zimmering et al, 1985], this compound was reported to be a potent mutagen when applied during larvae development [Kale et al, 1995]. It is in this context of conflicting data where we believe that our data from using the somatic mutation assay can be relevant, mainly taking into account that the quantification of the recombining activity of a compound is of primary importance for genotoxicity screening [Graf and Würgler, 1996].…”
