Mutational Analysis of the Cyanobacterial Nitrogen Regulator PipX

Abstract: PipX provides a functional link between the cyanobacterial global transcriptional regulator NtcA and the signal transduction protein PII, a protein found in all three domains of life as integrators of signals of the nitrogen and carbon balance. PipX, which is toxic in the absence of PII, can form alternative complexes with NtcA and PII and these interactions are respectively stimulated and inhibited by 2-oxoglutarate, providing a mechanism by which PII can modulate expression at the NtcA regulon. “…and pipX E4A alleles is counteracted by P II (17,19). To provide functional evidence that the mutant proteins PipX Y32A and PipX E4A exerted their effect on class 4 genes by interacting with NtcA rather than an NtcA-independent mechanism, we tested two functions that require NtcA in S. elongatus: growth on nitrate as a nitrogen source and nitrate transport.…”

“…In brief, two major targets of PII regulation have been identified at the molecular level, the transcriptional co-activator PipX and NAGK. PipX, a small protein of approximately 90 amino acids, was shown to be required for full expression of NtcA-activated genes (Espinosa et al 2006(Espinosa et al , 2007Laichoubi et al 2012). Binding of PipX to NtcA is favored by high 2-OG levels in vitro, whereas 2-OG in concert with Mg-ATP prevents binding of PipX to PII.…”

“…Under the physiological range of 2-OG levels, pipX mutations specifically impairing PipX-PII complexes favor formation of PipX-NtcA complexes. PipX residues Y32 and E4 are important for interactions with PII and NtcA proteins and Y32 for interactions with NtcA and these differences affect both NtcAdependent and independent genes [3,4] . The participation of PipX in at least 3 NtcA-independent scenarios was proposed.…”
