Mutations in the zebrafish hmgcs1 gene reveal a novel function for isoprenoids during red blood cell development

Abstract: Erythropoiesis is the process by which new red blood cells (RBCs) are formed and defects in this process can lead to anemia or thalassemia. The GATA1 transcription factor is an established mediator of RBC development. However, the upstream mechanisms that regulate the expression of GATA1 are not completely characterized. Cholesterol is 1 potential upstream mediator of GATA1 expression because previously published studies suggest that defects in cholesterol synthesis disrupt RBC differentiation. Here we charact… Show more

“…For each sh, a minimum of 20 to 30 z-stacks were collected. Maximum intensity projection images were processed and saved (32). For cell counts, the number of cells per zstack (20-30/ sh) at each jaw joint region (3 rows of chondrocytes on the left side of the joint and 5 rows on the right) were counted using ImageJ.…”

“…Polymerase chain reaction (PCR) was used to amplify the target sequence of the hmgcs1 gene. PCR and restriction digest was performed as previously described in (Quintana, Hernandez, & Gonzalez, 2017) (Hernandez et al, 2019).…”

“…Primers for qPCR are as follows: 1) Axin2 fwd 5’-CAACCAAGCACATCCATCAC-3’ and rev 5’-TCCATGTTCACCTCCTCTCC-3’, 2) rpl fwd 5’-TCCCAGCTGCTCTCAAGATT-3’ and rev 5’-TTCTTGGAATAGCGCAGCTT-3’, and 3) Sox10 fwd 5’-ACGCTACAGGTCAGAGTCAC-3’ and rev 5’-ATGTTGGCCATCACGTCATG-3’. Whole mount in situ hybridization was performed as previously described (Hernandez et al, 2019; Quintana et al, 2017; Thisse & Thisse, 2008). The axin2 riboprobe was produced with forward primer 5’-TGCACTGCTCCTTACATTCG-3’ and reverse primer 5’-GCTCCAGGACAAGGCTACTG-3’.…”

“…This region was selected because these cells were consistently visible across zstacks. Statistical signi cance was obtained by using a Student t-test with random control and morphant data (63). dlx2a expression is normal in morphants.…”
