MX1: a bending-magnet crystallography beamline serving both chemical and macromolecular crystallography communities at the Australian Synchrotron

Abstract: MX1 is a bending-magnet crystallography beamline at the 3 GeV Australian Synchrotron. The beamline delivers hard X-rays in the energy range from 8 to 18 keV to a focal spot at the sample position of 120 mm FWHM. The beamline endstation and ancillary equipment facilitate local and remote access for both chemical and biological macromolecular crystallography. Here, the design of the beamline and endstation are discussed. The beamline has enjoyed a full user program for the last seven years and scientific highlig… Show more

“…For PfA-M17 bound complexes, prior to data collection, crystals were soaked in mother liquor containing 1 mM ligand and 1 mM ZnSO 4 . Data were collected at 100 K using synchrotron radiation at the Australian Synchrotron using the macro crystallography MX1 beamline 3BM1 [32] for PfA-M1 and the micro crystallography MX2 beamline 3ID1 for PfA-M17. Diffraction images were processed and integrated using iMosflm [33] (PfA-M1) or XDS [34] (PfA-M17), scaled using Aimless [35] and solved by molecular replacement using Phaser [36] as part of the CCP4 suite [37].…”

“…The X-ray crystallography experiments were performed at the Australian Synchrotron, using silicon double crystal monochromated radiation at 100 K [40,41]. The crystal was rotated through Phi angle of 1-360 degrees.…”

“…The bond angles within the FeN 4 core range from 86.41 (8) to 156.79 (12) • with a dihedral angle between the almost planar six-membered chelating rings (rms deviation is 0.058 Å) of 81.36 (6) • . The smallest N-Fe-N angles are the intrachelate ones, whereas the largest one is that formed by the sulfonamide N atoms, i.e., N1-Fe1-N1A.…”

“…Recently commissioned beamlines dominantly or partly dedicated to small-molecule crystallography include, among others, I19 at the Diamond Light Source [6,7], MX1 at the Australian Synchrotron [8], and BL2D-SMC at the Pohang Light Source [9]. Examples of approved and very productive dedicated instruments include beamlines 11-3-1 [10] and 12-2-2 [11] at the Advanced Light Source (Berkeley, CA, USA).…”
