The aim of our study was to evaluate the effect of propolis on mortality of fish eggs due to mycosis, to study its efficacy on the fish growth rate and to analyze the histochemical and ultrastructural characteristics of muscle fibres. We evaluated the muscular development of fish in two different experimental conditions: traditional feeding and feeding with addition of propolis (groups A and B, respectively). The study was carried out on two species of teleosts bred in Italy, Oncorhynchus mykiss (rainbow trout) and Salmo trutta (brown trout). The use of propolis reduced mortality of fish eggs. The muscles were stained for myosin ATPase and succinic dehydrogenase to evaluate fibre type. The area, maximum and minimum diameter for each fibre type in relation to the body length were measured. The animals fed propolis showed a more rapid muscular growth compared to control fed the standard diet. Fibres were larger in the Salmo trutta than in the Oncorhynchus mykiss in both groups. In fish of A group, myotomal muscle, the presence of giant fibres and index of environmental stress was noticed. This type of study is valuable in order to extend the use of propolis as an antifungal agent and a natural product that can improve fish farming. But there are still many problems to solve, such as the support of ecocompatible activities, the improvement of animal health and welfare, product quality and consumer health assurance. Propolis is a mixture of various amounts of beeswax and resins collected by the honeybee from plants, particularly from flowers and leaf buds (Krell 1996). The colour of propolis ranges from yellow to dark brown depending on the origin of the resins. Propolis has attracted the researchers' interest in the last decades due to its biological and pharmacological properties, such as immunomodulatory, antitumor, antimicrobial, antiinflammatory, and antioxidant properties (Kanbur et al. 2009). The goal of this study was to analyze, for the first time, the influence of propolis on the fish muscular development and on the structural and ultrastructural characteristics of muscle fibre types. This study opens a new perspective on the investigation of biological properties of propolis, mainly with respect to the influence on the development of the trout and the mortality of fish eggs. The ability to utilize the RNA interference (RNAi) machinery for silencing target-gene expression has created a lot of excitement in the research community. In the present study, we used a cytomegalovirus (CMV) promoter-driven DNA template approach to induce short hairpin RNA (shRNA) triggered RNAi to block exogenous Enhanced Green Fluorescent Protein (EGFP) and endogenous No Tail (NTL) gene expressions. We constructed three plasmids, pCMV-EGFP-CMV-shGFP-SV40, pCMV-EGFP-CMV-shNTL-SV40, and pCMV-EGFP-CMV-shScrambled-SV40, each containing a CMV promoter driving an EGFP reporter cDNA and DNA coding for one shRNA under the control of another CMV promoter. The three shRNAgenerating plasmids and pCMV-EGFP control plasmid were introduced into zebrafish embryos by microinjection. Samples were collected at 48 h after injection. Results were evaluated by phenotype observation and real-time fluorescent quantitative reverse-transcription polymerase chain reaction (Q-PCR). The shGFP-generating plasmid significantly inhibited the EGFP expression viewed under fluorescent microscope and reduced by 70.05±1.26% of exogenous EGFP gene mRNA levels compared with controls by Q-PCR. The shRNA targeting endogenous NTL gene resulted in obvious NTL phenotype of 30±4% and decreased the level of their corresponding mRNAs up to 54.52 ± 2.05% compared with nontargeting control shRNA. These data proved the feasibility of the CMV promoter-driven shRNA expression technique to be used to inhibit exogenous and endogenous gene expressions in zebrafish in vivo.

Since its discovery, myostatin (MSTN) has been at the forefront of muscle therapy research because intrinsic mutations or inhibition of this protein, by either pharmacological or genetic means, result in muscle hypertrophy and hyperplasia. In addition to muscle growth, MSTN inhibition potentially disturbs connective tissue, leads to strength modulation, facilitates myoblast transplantation, promotes tissue regeneration, induces adipose tissue thermogenesis and increases muscle oxidative phenotype. It is also known that current advances in gene therapy have an impact on sports because of the illicit use of such methods. However, the adverse effects of these methods, their impact on athletic performance in humans and the means of detecting gene doping are as yet unknown. The aim of the present review is to discuss biosynthesis, genetic variants, pharmacological/genetic manipulation, doping and athletic performance in relation to the MSTN pathway. As will be concluded from the manuscript, MSTN emerges as a promising molecule for combating muscle wasting diseases and for triggering wide-ranging discussion in view of its possible use in gene doping. Skeletal muscle.Desde sua descoberta, a miostatina (MSTN) entrou na linha de frente em pesquisas relacionadas às terapias musculares porque mutações intrínsecas ou inibição desta proteína tanto por abordagens farmacológicas como genéticas resultam em hipertrofia muscular e hiperplasia. Além do aumento da massa muscular, a inibição de MSTN potencialmente prejudica o tecido conectivo, modula a força muscular, facilita o transplante de mioblastos, promove regeneração tecidual, induz termogênese no tecido adiposo e aumenta a oxidação na musculatura esquelética. É também sabido que os atuais avanços em terapia gênica têm uma relação com o esporte devido ao uso ilícito de tal método. Os efeitos adversos de tal abordagem, seus efeitos no desempenho de atletas e métodos para detectar doping genético são, contudo, desconhecidos. O objetivo da presente revisão de literatura foi discutir biossíntese, variantes genéticas, manipulação genética e farmacológica, e doping relacionado à via da MSTN. Como será concluído do manuscrito, a MSTN emerge como uma molécula promissora para combater doenças atróficas musculares e para gerar muitas discussões devido à sua possível utilização em doping genético.
