Growth differentiation factor-15 (GDF15), a member of the transforming growth factor-b superfamily, is associated with human cancer progress. We evaluated the role GDF15 plays in tumorigenesis of prostate carcinoma PC-3 cells. Results from real-time RT-PCR and ELISA revealed that expression of GDF15 was approximately threefold higher in LNCaP cells than in PC-3 cells. Other prostate cell lines (PZ-HPV-7, CA-HPV-10, and DU145 cells) expressed extremely low levels of GDF15. Stable overexpression of GDF15 in PC-3 cells enhanced the degree of cell proliferation and invasion as shown in the 3 H-thymidine incorporation assay and in the Matrigel invasion assay respectively. Soft agar assays and xenograft animal studies indicated that overexpression of GDF15 in PC-3 cells increased tumorigenesis in vitro and in vivo. Results from RT-PCR, immunoblot, and reporter assays revealed that overexpression of GDF15 resulted in decreased expression of maspin and upregulation of interleukin-6 (IL6), matriptase, and N-myc downstream-regulated gene 1 (NDRG1) expression. Further studies revealed that overexpression of IL6 enhanced GDF15 expression in LNCaP cells while knockdown of IL6 blocked the expression of GDF15 in PC-3 cells, suggesting that expression of GDF15 is upregulated by IL6. This study demonstrated that expression of GDF15 induces cell proliferation, invasion, and tumorigenesis of prostate carcinoma PC-3 cells. The enhancement of tumorigenesis and invasiveness of prostate carcinoma cells that stably overexpress GDF15 may be caused by the dysregulation of maspin, matriptase, and IL6 gene expression. The expression of GDF15 and IL6 is controlled via a positive feedback loop in PC-3 cells.

Abstract. the development of resistance to cisplatin during treatment of testicular cancer constitutes a major obstacle to the cure of testicular cancer. to resolve its mechanism will provide useful information for making clinical decisions. We found 4 and 15 gene expressions were, respectively, up-regulated and down-regulated in cisplatin-resistant testicular cancer nec-8/DDp cells compared with their parental nec-8 cells (about 2.5-fold) using cDnA microarray analysis. After screening, we selected the entpD6 among these 19 genes. entpD6 was less expressed in cancerous region compared with that in non-cancerous lesion. In addition, entpD6 expression in seminoma was higher than that in other testicular tumors. entpD6 expression was involved in cellular sensitivity to cisplatin through an interaction with e-cadherin. entpD6 is a promising molecular biomarker of cisplatin resistance in testicular cancer, and also a novel therapeutical target modulating cisplatin resistance in testicular cancer.

Abstract. Expression of n-myc downstream regulated gene 1 (ndrG1)/ca 2+ -associated protein 43 (cap43) in cancer cells is a predictive marker of good or poor prognosis depending on tumor type. in this study, we examined whether ndrG1/ cap43 is a marker of good or poor prognosis in gastric cancer patients, and whether it is associated with tumor stromal responses, including angiogenesis and macrophage infiltration. the expression levels of ndrG1/cap43, the number of cd68-positive macrophages and the cd34-positive microvessel density were analyzed by immunohistochemistry in 129 gastric cancer patients, including 65 with the intestinal type and 64 with the diffuse type. the expression of ndrG1/cap43 in the nucleus and the membrane was evaluated. nuclear ndrG1/ cap43 expression was found in 20/65 (30.8%) patients with the intestinal type and in 9/64 (14.1%) patients with the diffuse type of gastric cancer. nuclear ndrG1/cap43 expression was significantly associated with pathological stage in the intestinal type (p=0.002), but not in the diffuse type (p=0.039). nuclear ndrG1/cap43 expression was also closely associated with infiltrating macrophages (P=0.001) and tumor angiogenesis (p=0.001) in the intestinal type. Furthermore, nuclear ndrG1/cap43 expression was associated with poor prognosis in both the intestinal (p=0.001) and the diffuse types of gastric cancer (p=0.047).
