Na+/Ca2+ EXCHANGER INHIBITOR AMELIORATES IMPAIRED ENDOTHELIUM‐DEPENDENT Na+ RELAXATION INDUCED BY HIGH GLUCOSE IN RAT AORTA

Abstract: The present study was designed to investigate the effects of KB-R7943, an inhibitor of the Na+/Ca2+ exchanger, on impaired endothelium-dependent relaxation (EDR) induced by high glucose in rat isolated aorta. Both acetylcholine (ACh)-induced EDR and sodium nitroprusside (SNP)-induced endothelium-independent relaxation (EIR) were measured after aortic rings had been exposed to high glucose in the absence and presence of KB-R7943. Coincubation of aortic rings with high glucose (25 mmol/L) for 24 h resulted in a … Show more

“…Therefore, detailed investigations conducted in experimental diabetic models may provide clues to the mechanism of this dysfunction. Previous work from our laboratory suggests that KB-R7943 (an inhibitor of Na + /Ca 2 + exchanger) can restore impaired EDR induced by high glucose (or advanced glycosylation end products) in isolated rat aorta and exerted its beneficial effect by both inhibiting adhesion of monocytes to endothelial cells and the expression of intercellular adhesion molecule-1 in endothelial cells (Su et al, 2008;Li et al, 2010;Liu et al, 2015). However, there are no reports as to whether Na + /Ca 2 + exchanger inhibitor is able to protect against the impairment of endothelial functions in diabetic rat aorta.…”

“…KB-R7943 may have therapeutic potential as novel drugs for reperfusion injury and saltdependent hypertension and pulmonary hypertension (Poburko et al, 2007;Santo-Domingo et al, 2007;An et al, 2006;Iwamoto 2007). Previous work from our laboratory suggests that KB-R7943 can restore impaired endotheliumdependent relaxation induced by high glucose (or advanced glycosylation end products) in iso-lated rat aorta and KB-R7943 exerted its beneficial effect by both inhibiting adhesion of monocytes to endothelial cells and the expression of intercellular adhesion molecule-1 in endothelial cells (Su et al, 2013;Su et al, 2008;Li et al, 2010). This study was designed to investigate the effects of KB-R7943 on NCX expression, NCX activity and oxidative stress index in high glucose condition.…”
