NAMPT and BMAL1 Are Independently Involved in the Palmitate-Mediated Induction of Neuroinflammation in Hypothalamic Neurons

Abstract: Obesity is a prominent metabolic disease that predisposes individuals to multiple comorbidities, including type 2 diabetes mellitus, cardiovascular diseases, and cancer. Elevated circulating levels of fatty acids contribute to the development of obesity, in part, by targeting the hypothalamus. Palmitate, the most abundant circulating saturated fatty acid, has been demonstrated to dysregulate NAMPT and circadian clock proteins, as well as induce neuroinflammation. These effects ultimately result in hypothalamic… Show more

“…Diet is also capable of remodeling circadian rhythms behaviorally, transcriptionally, and metabolically [ 22 , 260 , 272 , 273 ]. For example, feeding mice a ketogenic diet or overexpressing the FGF21 (whose production is stimulated by a ketogenic diet) augments locomotion during the daytime and results in a phase advance in mice [ 183 ].…”

“…For example, feeding mice a ketogenic diet or overexpressing the FGF21 (whose production is stimulated by a ketogenic diet) augments locomotion during the daytime and results in a phase advance in mice [ 183 ]. Mice placed on standard HFD have a profound reorganization of their circadian gene expression and metabolite production, in several metabolic tissues, including liver, adipose tissue, and the CNS [ 22 , 272 , 273 ]. However, hypothalamic PER2::LUC rhythms in the SCN, DMH or ARC appear unaltered under HFD [ 91 ].…”

“…Thus, the increases in have been inconsistent (Busso et al, 2008;Revollo et al, 2007). We reported that inhibiting NAMPT activity with FK866 prevented the palmitate-mediated induction of Il-6 and Tlr4 mRNA, suggesting a role for this enzyme in mediating neuroinflammation (Tran et al, 2020).…”

“…Thus, increased visfatin may have deleterious effects on energy homeostasis. In fact, visfatin induces insulin resistance in the liver partly via induction of inflammatory pathways and induces FA-mediated neuroinflammation ( 152 , 153 ).…”
