Nanoengineering of Stimuli‐Responsive Protein‐Based Biomimetic Protocells as Versatile Drug Delivery Tools

Abstract: We present a general strategy to nanoengineer protein-based colloidal spheres (biomimetic protocells) as versatile delivery carriers with stimuli responsiveness by the electrostatic assembly of binary components (proteins and polypeptides) in association with intermolecular disulfide cross-linking. The size of the colloidal spheres, ranging from nanoscale to microscale, is readily tuned through parameters like protein and polypeptide concentration, the ratio between both, pH, and so on. Moreover, such colloida… Show more

“…In addition, the cells showed marked red fluorescence after uptake BPHM in cancer cells, which makes the compound a potential agent for medical fluorescence imaging. Considering that BPHM can rapidly enter cells, and possessed the hydrophobic character due to the phenylhydrazine structure, hence it may agglomerate when subjected to aqueous solution and may enters cells at the nano-scale [30][31][32]. In order to investigate how BPHM enters cells (in molecular states or nanoparticles), we performed dynamic light scattering (DLS) measurement in phosphate buffer saline.…”

“…[12] Basically, the crosslinking was mainly performed by heating the PEM films or using chemical crosslinkers, which causes the reduction of the amount of incorporated entities or longer crosslinking time. [13][14][15][16][17][18][19] More recently the crosslinking with photo-active molecules was introduced, which allows a drastically decreased crosslinking time, whereby defined patterns for tuning optical properties or an increased film rigidity for cell adsorption was created. [20,21] One of the great advantages of this method is to avoid poisonous chemicals like glutaraldehyde.…”
