Nanomedicine Strategies for Heating “Cold” Ovarian Cancer (OC): Next Evolution in Immunotherapy of OC

Abstract: Immunotherapy has revolutionized cancer treatment, dramatically improving survival rates of melanoma and lung cancer patients. Nevertheless, immunotherapy is almost ineffective against ovarian cancer (OC) due to its cold tumor immune microenvironment (TIM). Many traditional medications aimed at remodeling TIM are often associated with severe systemic toxicity, require frequent dosing, and show only modest clinical efficacy. In recent years, emerging nanomedicines have demonstrated extraordinary immunotherapeut… Show more

“…However, most conventional antioxidants are usually confined by the poor dispersity, low stability, and short duration of action. Meanwhile, natural enzymes and single-component nanozymes usually show a high specificity for a certain ROS and therefore, fail to scavenge various ROS generated during disease progression ( Goyal et al, 2014 ; Yang et al, 2022a ; Yang et al, 2022b ; Xiao et al, 2022 ). Furthermore, the synthesis of the multienzyme-based antioxidant is often complicated, shows low stability, and is difficult to be repeated.…”

“…Ovarian cancer with low tumor-infiltrating lymphocyte is considered “cold” tumor ( Yang et al, 2022 ). The results of our immune infiltration analysis also showed no difference in activated CD8 T-cells between the high and low risk score subgroups.…”

“…High levels of RONS disrupt vital cellular processes in many oxidative stress related diseases by attacking DNA due to their high reactivity [ 76 , 79 , 173 ]. Unfortunately, unlike tumor tissues which have developed efficient mechanisms of RONS detoxification (mainly involving enhancement of antioxidant abilities and upregulation of pro-survival and pro-repair molecules), normal tissues such as kidneys are difficult to cope with large-scale oxidative stress in time [ 174 , 175 ]. Correspondingly, the administration of exogenous accessible DNA can directly react with RONS to reduce cell damage and alleviate AKI.…”
