Nanomolar detection of L-cysteine and Cu2+ ions based on Trehalose capped silver nanoparticles

In current scenario, plant-mediated synthesis of nanoparticles has received significant interest owing to its cost-effectiveness and eco-friendly methods. This study was planned to find the potential of Carduus edelbergii Rech.f. extract for the fabrication of silver nanoparticles (AgNPs) and their subsequent antibacterial, antifungal, cytotoxic, and antioxidant activities. The prepared plant extract was analyzed for phytochemicals, elements, and nutrients. The fabricated AgNPs were characterized by spectroscopy (UV-Vis), X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FTIR), and scanning electron microscopy (SEM). The synthesized AgNPs and the Carduus edelbergii extract were investigated for their antioxidant, antibacterial, cytotoxic, and antifungal characteristics. The results showed that the plant extract contains a wide range of important nutrients and secondary metabolites. The synthesized AgNPs were crystalline and globular with an average diameter of 36.5 nm and exhibited a localized surface plasmon resonance (LSPR) peak at roughly 420 nm. As compared to Carduus edelbergii extract, the phytofabricated AgNPs had an increased antioxidant and cytotoxic efficacy. The AgNPs showed substantial antibacterial action against Gram-positive (Staphylococcus epidermis and Staphylococcus aureus) and Gram-negative (Klebsiella aerogenes and Escherichia coli) bacteria. The findings revealed that the phytofabricated AgNPs could be an effective antioxidant, antibacterial, and antifungal for treatment of medical conditions.

Following access into the cell, colloidal silver nanoparticles exhibit generalized cytotoxic properties, thus appear as omnipotent microbicidal, but not suitable for systemic use unless are free of toxic effects on host cells. The AgNP-Serum-18 when prepared from silver nitrate, using dextrose as reducing and group-matched homologous serum as a stabilizing agent, selective endocytosis, and oxidative stress-dependent bio-functional damages to the host are mostly eliminated. For their bio-mimicking outer coat, there is the least possibility of internalization into host cells or liberation of excess oxidants in circulation following interaction with erythrocytes or vascular endothelial cells. The presence of infection-specific antibodies in the serum can make such nano-conjugates more selective. A potent antimicrobial action and a wide margin of safety for mammalian cells in comparison with very similar PVA-capped silver nanoparticles have been demonstrated by the in-vitro challenge of such nanoparticles on different microbes, human liver cell-line, and in-vivo study on mice model. This may open up wide-range therapeutic prospects of colloidal nanoparticles.

Inducing autophagy of macrophages to improve abnormal lipid metabolism is an important way to treat atherosclerosis (AS). Yet, the current application of the mammalian target of rapamycin (mTOR)-dependent autophagy inducers is limited by the side effects and lack of targeting and low biological availability. Herein, a kind of nitric oxide (NO)-driven carrier-free nanomotor based on the reaction between trehalose (Tr, one of the mTOR-independent autophagy inducers), L-arginine (Arg), and phosphatidylserine (PS) is reported. The developed nanomotors use NO as the driving force, which is generated from the reaction between Arg and excessive reactive oxygen species (ROS) and inducible nitric oxide synthase (iNOS) specifically presenting in the AS microenvironment. The high expression of ROS and iNOS in the AS site can be used as chemoattractants to induce chemotaxis behavior of the nanomotors to achieve the first-step targeting an AS plaque. Subsequently, the “eat me” signal sent by PS is exploited to precisely target to the macrophages in the AS plaque, realizing the plaque-macrophage-targeted effect by this step-by-step strategy. In vitro and in vivo results confirm that the introduction of the concept of carrier-free nanomotors has greatly improved the biological availability of trehalose (the dose can be reduced from 2.5 g kg–1 in previous reports to 0.01 g kg–1 in this work). Particularly, consumed ROS and the production of NO during the targeting process also play positive roles, in which the former regulates the M2 polarization of macrophages and the latter promotes the reconstruction of an endothelial barrier, which contributes to the multilink treatment of AS.
