Nanoscale Porphyrin Metal-Organic Frameworks Deliver siRNA for Alleviating Early Pulmonary Fibrosis in Acute Lung Injury

Abstract: Acute lung injury (ALI) has high mortality and still lacks novel and efficient therapies. Zinc finger E-box binding homeobox 1 and 2 (ZEB1/2) are highly expressed in the early stage of ALI and are positively correlated with the progression of pulmonary fibrosis. Herein, we developed a nanoscale Zr(IV)-based porphyrin metal-organic (ZPM) framework to deliver small interfering ZEB1/2 (siZEB1/2) to alleviate early pulmonary fibrosis during ALI. This pH-responsive nano-ZPM system could effectively protect siRNAs d… Show more

“… 637 Additionally, ALI can develop into acute respiratory distress syndrome in more serious injuries, which lacks novel and efficient therapies. 638 , 639 Inflammation and oxidative stresses are essential for the progression of ALI. 640 However, the molecular mechanisms of sepsis-induced lung inflammatory injury are yet to be determined.…”

“…In order to find a way to alleviate early pulmonary fibrosis in mice, Weng et al. prepared a nanoparticle ZPM@PDE‐siZEB1/2, [88] which delivered siZEB1 and siZEB2 into the lungs of ALI mice through trachea administration. Compared with the lung tissues of mice in the control group and LPS group, the symptoms of LPS+ZPM@PDE‐siZEB1/2 group were significantly relieved, the expression levels of ZEB1 and ZEB2 are significantly decreased.…”
