Nap1: taking a closer look at a juggler protein of extraordinary skills

Abstract: The nucleosome assembly protein Nap1 is used extensively in the chromatin field to reconstitute nucleosomal templates for structural and functional studies. Beyond its role in facilitating experimental investigation of nucleosomes, the highly conserved Nap1 is one of the best-studied members of the histone chaperone group. Here we review its numerous functions, focusing mainly on its roles in assembly and disassembly of the nucleosome particle, and its interactions with chromatin remodeling factors. Its presum… Show more

“…In more physiological buffers, further oligomerization of yNAP was observed using size exclusion chromatography (SEC) (12) and AUC (25). Several groups reported that the NAP1 dimer can bind an H3/H4 or H2A/H2B dimer or an H1 monomer (11,12,25,26). Alternative stoichiometries have been suggested, including one NAP1 monomer binding one histone dimer (27,28), and therefore, the question of the NAP1-histone stoichiometry remains controversial.…”

“…To complete nucleosome formation following the delivery of two dimers of H3 -H4 onto newly synthesized DNA, the subsequent addition of histones H2A-H2B involves the nucleosome assembly protein 1 (NAP1) chaperone (Zlatanova et al 2007). Given that the FACT (facilitates chromatin transcription) complex also acts as an H2A-H2B chaperone in transcription, DNA replication, and DNA repair, it could perhaps help to provide a connection with NAP1 (Krogan et al 2006).…”
