Naphthyl-Poly(S-((2-carboxyethyl)thio)-l-cysteine) Peptide Amphiphiles with Different Degrees of Polymerization: Synthesis, Self-Assembly, pH/Reduction-Triggered Drug Release, and Cytotoxicity

Abstract: Four peptide amphiphiles (PA1-4) with different degrees of polymerization (DP = 40,15,10,and 6) were synthesized by Fuchs-Farthing and ring-opening polymerization followed by post-polymerization modification, as fully characterized by 1 H NMR, FT-IR, gel permeation chromatography, and circular dichroism (CD) spectroscopy. It was found that PAs could self-assemble to form regular spherical micelles in lowconcentration (about 1 mg/mL) aqueous solution, which had different contents of secondary structures and mai… Show more

“…When GSH was added, these values increased to 64%, 67% and 72%, indicating that the synthesized hydrogel was pH/redox dual-responsive. 88 Wang et al designed an in situ self-assembly system (LND-GFFYK(TPP)-ss-PEG) that targets mitochondria to induce mitochondrial dysfunction and autophagy by releasing mitochondrial-targeting peptides and loaded-drugs upon cleavage by GSH. 89 …”

“…Micelles provide a way for drugs to bypass the cell barrier, which they normally cannot do (Azum et al, 2021; Basu et al, 2023; Behari et al, 2022; Schittny et al, 2020). Micelles are a promising drug delivery method due to their many advantages in the areas of size, toxicity, drug solubility, and cytotoxicity regulation (Negut & Bita, 2023; Xu et al, 2023). Due to the synergistic effects of a mixed surfactant solution, mixed surfactant systems perform better than single surfactant systems and are often utilized in formulations for many applications in several industries.…”
