The COP9 signalosome (CSN) is an evolutionarily conserved multi-subunit protein complex, which controls protein degradation through deneddylation and inactivation of cullin-RING ubiquitin E3 ligases (CRLs). Recently, the CSN complex has been linked to the NF-κB signaling pathway due to its association with the IKK complex. However, how the CSN complex is regulated in this signaling pathway remains unclear. Here, we have carried out biochemical experiments and confirmed the interaction between the CSN and IKK complexes. In addition, we have determined that overexpression of IKKα or IKKβ leads to enhanced phosphorylation of CSN5, the catalytic subunit for CSN deneddylase activity. Mutational analyses have revealed that phosphorylation at serine 201 and threonine 205 of CSN5 impairs CSN-mediated deneddylation activity in vitro. Interestingly, TNF-α treatment not only enhances the interaction between CSN and IKK, but also induces an IKK-dependent phosphorylation of CSN5 at serine 201, linking CSN to TNF-α signaling through IKK. Moreover, TNF-α treatment affects CSN interaction network globally, especially the associations of CSN with the proteasome complex, eukaryotic translation

Over the last three decades, technology companies have promoted the "disruptive" potentials of the information age. Brazilian favelas (shantytowns) provide one of the most popular examples used to describe how digital disruption applies to informal urban communities. Favelas have been mapped by Google and surveilled by an IBM Smart City while hundreds of well-branded digital inclusion programs present themselves as alternatives to an informal economy and an illicit drug trade. However, corporate narratives of digital disruption fail to account for what scholars describe as an "insurgency" and practices of improvisation (gato, jeitinho, or gambiarra) found in the favela. Describing a process of "converting" regulatory fines into well-branded social projects, this article provides an ethnographic account of a Microsoft-funded documentary about an ex-drug trafficker turned digital educator. Considering the role of ethnography in an urban "gray zone," this article asks: what techniques do global technology corporations use to take symbolic ownership of local knowledge? What does the dissonance between corporate and community-based narratives reveal about alternative forms of creativity in the digital age? And, how can we characterize the formalizing potentials of digital disruption in Latin America? Goffman and the Mafia: shaping YouTube's technological affordances in the war on drugs

This paper advances the discussion about the interactions enabled through communication technologies by articulating Goffman's theory of strategic interactions and Trevor Pinch's concept of copresence, and applying them to analyze the way a Mafia's armed wing posted videos on YouTube during the turf war developed in Ciudad Juarez, Mexico, from 2008 to 2011. I analyzed the videos and the comments set up below them. Science and Technology Studies scholars have been engaged in a debate about how to study online contents. They agree that STS should advance approaches that only give an account of the semiotic properties of the contents, in order to explore the interactions they enable. However, they don't agree on how this should be done. This study lends support to perspectives arguing that Goffman's theory is still relevant to analyze online interactions, in spite of having written his theory before the expansion of the Internet. I provide examples to argue that, in fact, Goffman was fully aware that interactions could be technologically mediated. I suggest that members of the armed wing shaped YouTube's technological affordances to make themselves accountable to other parties in the war, and available for interaction.
