ABSTRACTAround the world, there are numerous alkaline, hypersaline environments that are heated either geothermally or through intense solar radiation. It was once thought that such harsh environments were inhospitable and incapable of supporting a variety of life. However, numerous culture-dependent and -independent studies revealed the presence of an extensive diversity of aerobic and anaerobic bacteria and archaea that survive and grow under these multiple harsh conditions. This diversity includes the halophilic alkalithermophiles, a novel group of polyextremophiles that require for growth and proliferation the multiple extremes of high salinity, alkaline pH, and elevated temperature. Life under these conditions undoubtedly involves the development of unique physiological characteristics, phenotypic properties, and adaptive mechanisms that enable control of membrane permeability, control of intracellular osmotic balance, and stability of the cell wall, intracellular proteins, and other cellular constituents. This minireview highlights the ecology and growth characteristics of the extremely halophilic alkalithermophiles that have been isolated thus far. Biochemical, metabolic, and physiological properties of the extremely halophilic alkalithermophiles are described, and their roles in resistance to the combined stressors of high salinity, alkaline pH, and high temperature are discussed. The isolation of halophilic alkalithermophiles broadens the physicochemical boundaries for life and extends the boundaries for the combinations of the maximum salinity, pH, and temperature that can support microbial growth.

Halovivax asiaticus gen. nov., sp. nov., a novel extremely halophilic archaeon isolated from Inner Mongolia, China Strain EJ-46 T , a novel pleomorphic, aerobic, extremely halophilic member of the Archaea was isolated from sediment of the saline Lake Ejinor, in Inner Mongolia, China. This organism was neutrophilic and required at least 15 % (2?5 M) NaCl for growth. MgCl 2 was not required. The isolate was able to grow at pH 6?0-9?0. Optimum growth occurred in media containing 20 % (3?4 M) NaCl at pH 7?0-7?5. Polar lipid analysis revealed the presence of phosphatidylglycerol and phosphatidylglycerol phosphate methyl ester, derived from both C 20 C 20 and C 20 C 25 glycerol diethers. Four glycolipids were detected, one of which may be novel. The DNA G+C content was 60?3 mol%. 16S rRNA gene analysis revealed that strain EJ-46 T was a member of the phylogenetic group defined by the family Halobacteriaceae, and the highest 16S rRNA gene similarity values of 94?9 and 94?8 % were obtained with the haloalkaliphilic species of the genus Natronococcus, Natronococcus occultus and Natronococcus amylolyticus, respectively. Based on the phenotypic, genotypic and phylogenetic analyses, it is proposed that the novel isolate should be classified as representing a new genus and species, for which the name Halovivax asiaticus gen. nov., sp. nov. is proposed. The type strain is EJ-46 T (=CGMCC 1.4248 T =CECT 7098 T ).The extremely halophilic, aerobic members of the Archaea are classified within the family Halobacteriaceae, order Halobacteriales, with 20 currently recognized genera: Halalkalicoccus Members of the Halobacteriaceae are usually pink-to redpigmented because of the presence of carotenoids (Kamekura & Dyall-Smith, 1995;Hezayen et al., 2001;Grant et al., 2001). They have been isolated from various hypersaline environments, such as saline lakes (e.g. Franzmann et al., 1994;Norton et al., 1993). The haloarchaea are the most halophilic organisms known and are dominant in hypersaline environments in which the salt concentration exceeds 25 % (w/v) (Rodriguez-Valera et al., 1981). They often appear at such high density that they impart a typical red colour to the hypersaline brines.Hypersaline environments are commonly found in China. In addition to many coastal salterns, a number of salt lakes, soda lakes and salt-rich deserts are located in various geographical areas of China, e.g. Xinjiang, Inner Mongolia and Tibet Autonomous Regions. The present study deals with the isolation and characterization of the moderately halophilic-alkaliphilic bacteria from a saline habitat in western India. Eight different bacterial strains were isolated using enrichment techniques at 20% (w/v) NaCl and pH 10. The isolates exhibited diversity towards gram ' s reaction, colony and cell morphology. They were able to grow and produce alkaline protease over a broad range of NaCl, 5-20% (w/v) and pH, 8-10. None of the isolates could grow at pH 7, and one could not grow even at pH 8. Crude and partially purified proteases from strain S5 were subjected to characterization with reference to pH, salt stability and protein folding. Optimum protease activity and stability was recorded at 10% salt and pH 9-9.5. Denaturation kinetics of S5 alkaline protease along with a reference protease was studied at 8M urea followed by renaturation. The S5 alkaline protease could be partially renatured up to 32% of the original activity. Despite of the fact that all the 8 isolates were from the same site, they displayed significant diversity with respect to their salt requirement for growth and enzyme secretion. While the effect of pH was less demarcated on growth, the protease production was significantly affected. Isolate S 5 produced substantial amount of halotolerant and alkaline protease. The activity and stability of the alkaline protease in a broader range of pH and salt would definitely make this enzyme an important candidate for various industrial applications.
