Natural chaperonin of the hyperthermophilic archaeum, Thermococcus strain KS-1: a hetero-oligomeric chaperonin with variable subunit composition

Abstract: SummaryTo study the difference in expression of the chaperonin a-and b-subunits in Thermococcus strain KS-1 (T. KS-1), we measured their intracellular contents at various growth temperatures using subunit-specific antibodies. The b-subunit was significantly more abundant with increasing temperature (maximum at 938C), whereas the a-subunit was not. Native PAGE with Western blot analysis indicated that the natural chaperonins in the crude extracts of T. KS-1 cells grown between 658C and 958C migrate as single ba… Show more

“…Because of the high sequence homology between TkCPN and PhCPN, the anti-TkCPN antibody can specifically immunoprecipitate PhCPN. The preparation and purification of the antibody were described previously (29). The sample solutions for the immunoprecipitation containing 7.0 M PhPFD and 10 mM HCl-unfolded GFP (at the specified concentration) were mixed and incubated at 65°C for 10 min prior to the addition of 1.4 M PhCPN.…”

“…strain KS-1. The natural chaperonin derived from T. KS-1 is a hetero-oligomer of ␣ and ␤ subunits, and the composition varies with the growth temperature (14). In addition, each subunit forms a double ring homo-oligomer capable of ATP-dependent protein folding in vitro (10).…”

“…strain KS-1 (T. KS-1), is one of the most studied group II chaperonins (17)(18)(19)(20)(21)(22)(23). T. KS-1 chaperonin is composed of two highly homologous subunits, ␣ and ␤.…”

“…T. KS-1 chaperonin is composed of two highly homologous subunits, ␣ and ␤. Although the natural chaperonin isolated from T. KS-1 is a hetero-oligomer, each of the recombinant ␣ and ␤ subunits forms a double-ring homo-oligomer and functions as a chaperonin in vitro (17)(18)(19). Recent experimental results using ␤-chaperonin and natural chaperonin suggest that the protein folding occurs within the cis-cavity (21).…”
