Natural killer cells: walking three paths down memory lane

Abstract: Immunological memory has traditionally been regarded as a unique feature of the adaptive immune response, mediated in an antigen-specific manner by T and B lymphocytes. All other hematopoietic cells, including natural killer (NK) cells, are classified as innate immune cells, which have been considered short-lived but can respond rapidly against pathogens in a manner not thought to be driven by antigen. Interestingly, NK cells have recently been shown to survive long term after antigen exposure and subsequently… Show more

“…Recent studies from several groups have highlighted the existence of long-lived NK cells that display characteristics of immunological memory, including more robust responsiveness upon secondary stimulation (33). Such "memory" NK cells have been postulated to contribute to immunity in the context of cytomegalovirus infections in both mice (34) and humans (35).…”

“…T-cell epitopes encoded by LCMV included NP 396 -404 (FQ PQNGQFI), GP [33][34][35][36][37][38][39][40][41] (KAVYNFATC), GP 61-80 (GLKGPDIYKGVYQFK SVEFD), and GP 276 -286 (SGVENPGGYCL). Peptides were purchased from 21st Century Biochemicals and purified by reverse-phase high-pressure liquid chromatography (HPLC) to 90% purity.…”

“…On the other, they secrete classic lymphocyte cytokines and in some cases exhibit classic memory. Among innate lymphoid cells, natural killer cells pose the biggest classification challenge because they are capable of interacting with MHC molecules and generating antigenspecific immunologic memory that mirrors that of adaptive T lymphocytes (29). The precise role that innate lymphoid cells have in immunity in general and in kidney disease in particular remains to be determined.…”

“…The term memory-like NK cells, which means NK cells similar to memory cells, is used in literature. Some scientists point out three possible variants of memory-like NK cells, which differ by mechanism of formation and functional characteristics: virus dependent induction, cytokine induction, and existence of specialized population of memory-like NK cells in liver [24]. A capacity of NK cells, previously activated by IL-12 and IL-18 cytokines, after a certain period of time (from 7 to 21 days) at repeated cytokine stimulation to produce an increased number of IFNγ compared to control was described [30].…”
