NBLAST: Rapid, sensitive comparison of neuronal structure and construction of neuron family databases

Abstract: Neural circuit mapping is generating datasets of 10,000s of labeled neurons. New computational tools are needed to search and organize these data. We present NBLAST, a sensitive and rapid algorithm, for measuring pairwise neuronal similarity. NBLAST considers both position and local geometry, decomposing neurons into short segments; matched segments are scored using a probabilistic scoring matrix defined by statistics of matches and non-matches.We validated NBLAST on a published dataset of 16,129 single Drosop… Show more

“…This effort uses serial-section electron microscopy combined with cell-type specific genetic tools to map synaptic connections between morphologically identifiable cell types [204, 205]. Cell types identified morphologically can then be matched with driver lines using new bioinformatic tools [206]. This effort to map synaptic connections using large-scale electron microscopy has focused on the brain thus far, but it is currently being extended to the VNC as well.…”

“…All annotated synapses in this wiring diagram fulfill the four following criteria of mature synapses In this study we validate the reconstructions as previously described 11;55 , a method successfully employed in multiple studies 11;55-60 . Briefly, in Drosophila, as in other insects, the gross morphology of many neurons is stereotyped and individual neurons are uniquely identifiable based on morphology [61][62][63] . Furthermore, the nervous system in insects is largely bilaterally symmetric and homologous neurons are reproducibly found on the left and the right side of the animal.…”

“…9, Supplementary Table 1, and Supplementary Data 1 and 2 ), PACT and PARS provide a means for efficiently acquiring information on the spatial position of neurons within large tissue volumes at high resolution. For this information to be applied to mapping the connectome 59 , however, these gigabytes or even terabytes of raw image data (e.g., for a whole mouse brain at 25× magnification) must be converted into a complex network of neuron projection pathways and neural contacts, a feat that poses substantial demands on both storage hardware and image analysis software. Many available software tools and image file formats were not designed with tera-scale data sets in mind and assume that entire image volumes fit in computer RAM.…”
