NDP kinase moves into developing primary cilia

Abstract: 3 SUMMARYInmunofluorescence staining of murine NIH3T3 fibroblasts grown at high density shows that conventional nucleoside diphosphate (NDP) kinases A and B localize to a sensory organelle, the primary cilium. Similar results are obtained with Xenopus A6 kidney epithelial cells, suggesting that NDP kinases are a universal component of the primary cilium. The translocation of NDP kinase into primary cilia depends on size, taking place only when cilia reach a critical length of 5-6 μm. In mature cilia, NDP kinas… Show more

“…If this is to be investigated in cells in culture or tissues by means other than, or in addition to, fluorescence microscopy, then it would be helpful to have efficient methods for the isolation of primary cilia and, in turn, ciliary membranes from these cells. Although methods for doing this have been published, it is technically demanding, and only small amounts of cilia are obtained [51,52]. …”

“…Just recently, these two thioredoxins have been involved in the control of flagellar motility through their interactions with flagellar components (41). In addition, from Chlamydomonas to mammals, NDP kinases are also found in ciliary structures where they are supposed to play essential functions, particularly through their ability to produce GTP (42)(43)(44). Taken together, these observations strongly support ., pTXNDC3C 1, pTXNDC3C2, pTXNDC3T1ϾC, and pTXNDC3T2ϾC) or a thymine at that position (i.e., pTXNDC3T 1, pTXNDC3T2, pTXNDC3C1ϾT, and pTXNDC3C 2ϾT) (see Materials and Methods).…”
