Near-Infrared Dye-Conjugated Amphiphilic Hyaluronic Acid Derivatives as a Dual Contrast Agent for In Vivo Optical and Photoacoustic Tumor Imaging

Abstract: Amphiphilic hyaluronic acid (HA) derivatives bearing hydrophobic indocyanine green dye derivatives and hydrophilic poly(ethylene glycol) were synthesized through the use of condensation and copper-catalyzed click cyclization reactions. The amphiphilic HA derivatives dissolved in water and formed self-assemblies in which the near-infrared dyes were tightly packed and arranged to form dimers or H-aggregates. By irradiating an aqueous solution of HA derivatives with near-infrared light, photoacoustic signals were… Show more

“…A recent report demonstrates that ICG that was directly conjugated to poly(ethylene glycol) grafted-HLA, where ICG served as the hydrophobic ligand, resulted in tumor signal, both fluorescence and photoacoustic, that was more than double that of femur. 54 However, the dose of ICG used in this study, 50 nmol per mouse, was 5 times greater than the dose used in this study.…”

“…By utilizing HA derivatives as contrast agents, tumors were clearly visualized by optical imaging as well as by OA tomography. 52 Because skin is composed of alternating lipophilic and hydrophilic layers, amphipathic compounds that possess both properties may be absorbed by the skin more effectively. Hydration is the most widely used and safest method to increase skin penetration for both hydrophilic and lipophilic permeants.…”

“…HA derivatives bearing both hydrophobic (indocyanine green) and hydrophilic (PEG) components were developed, but these substances did not possess the capability of enhancing penetration of light through skin. 52 To transport drugs effectively through the cellular membrane and deliver them into the intracellular environment, several smart carrier systems based on HA composites were developed containing both synthetic or natural polymers. 56 We speculate that the method of HA pretreatment described in this work may be also effective for drug delivery.…”

“…Fifth, the use of VFR has not been validated in the infrapopliteal artery in the clinical setting, although we have recently reported the ability for dilating the vascular bed exists in the lower limb during increased oxygen demand supply in response to a given hyperemic stimulation and can be assessed in the same manner. 21 Finally, the pressure measurement distal to the lesion immediately after EVT was not attempted because residual dissections could be extended by advancing the pressure sensor into the artery undergoing EVT.…”
