Nematicidal Characterization of Solanum nigrum and Mentha arvensis Leaf Extracts Using Caenorhabditis elegans as a Model Organism

Considering foremost global issues instigated by parasitic nematodes, Solanum nigrum (S. nigrum) and Mentha arvensis (M. arvensis) nematicidal potential at the gene level has been explored herein. Methanol, ethyl acetate, chloroform, n-hexane, and distilled water were used for extract preparation. Caenorhabditis elegans (C. elegans) was used as the model organism. Nematicidal and anti-egg hatching assays, fluorescence microscopy, and quantitative real-time PCR were done. S. nigrum chloroform (LD 50 = 1.21 mg/mL) and M. arvensis methanol (LD 50 = 2.47 mg/mL) extracts exhibited excellent nematicidal potential. Both plants showed potent anti-egg hatching activity (1 mg/mL). S. nigrum methanol and M. arvensis ethyl acetate extracts showed high apoptotic effect in muscles, gonads, and uterus (eggs). Stress genes, that is, gst-4, hsp-16.2, and gpdh-1 were highly expressed in affected C. elegans (treated with S. nigrum and M. arvensis leaf extracts) when compared with normal C. elegans. Phytochemicals and bioactive compounds present in plants may be the major cause of their excellent nematicidal potential, which further confirmed that both plants could be an alternative candidate(s) for novel broad-scale anthelmintic drug(s).

Aging is a natural phenomenon that occurs in all living organisms. In humans, aging is associated with lowered overall functioning and increased mortality out of the risk for various age-related diseases. Hence, researchers are pushed to find effective natural interventions that can promote healthy aging and extend lifespan. Royal jelly (RJ) is a natural product that is fed to bee queens throughout their entire life. Thanks to RJ, bee queens enjoy an excellent reproductive function and lengthened lifespan compared with bee workers, despite the fact that they have the same genome. This review aimed to investigate the effect of RJ and/or its components on lifespan/healthspan in various species by evaluating the most relevant studies. Moreover, we briefly discussed the positive effects of RJ on health maintenance and age-related disorders in humans. Whenever possible, we explored the metabolic, molecular, and cellular mechanisms through which RJ can modulate age-related mechanisms to extend lifespan. RJ and its ingredients—proteins and their derivatives e.g., royalactin; lipids e.g., 10-hydroxydecenoic acid; and vitamins e.g., pantothenic acid—improved healthspan and extended lifespan in worker honeybees Apis mellifera, Drosophila Melanogaster flies, Gryllus bimaculatus crickets, silkworms, Caenorhabditis elegans nematodes, and mice. The longevity effect was attained via various mechanisms: downregulation of insulin-like growth factors and targeting of rapamycin, upregulation of the epidermal growth factor signaling, dietary restriction, and enhancement of antioxidative capacity. RJ and its protein and lipid ingredients have the potential to extend lifespan in various creatures and prevent senescence of human tissues in cell cultures. These findings pave the way to inventing specific RJ anti-aging drugs. However, much work is needed to understand the effect of RJ interactions with microbiome, diet, activity level, gender, and other genetic variation factors that affect healthspan and longevity.

Increasing evidence suggests that royal jelly (RJ) has exceptional biological properties, and that major royal jelly proteins (MRJPs) are the key active factors in RJ. The objective of this study was to compare the difference in the protein content between RJ and MRJPs using non-labeled, quantitative proteomics technology, and to investigate the adjustment features and mechanisms of MRJPs on murine immune functions and the composition of intestinal flora in cyclophosphamide-treated mice. Results showed that, during the process of extracting MRJPs, the ratio of the protein types in the main protein and other proteins decreased significantly, except for MRJP1 and MRJP7, which demonstrated that an enriching effect of MRJP1 and MRJP7 was present during the extraction process. Cyclophosphamide-induced mice were orally administered MRJPs. Results showed that the middle-dose group, which received 0.25 g/(kg·bw) of royal jelly main protein, demonstrated a clear impact on the development of the spleen and liver, the quantity of peripheral blood leukocytes, immunoglobulin content, immune factor level, and the proliferation ability of spleen lymphocytes. A 16S rRNA high-throughput sequencing technology analysis showed that MRJPs could improve the component and richness of intestinal flora and raise the immunity of mice. The above-mentioned results indicated that the application of MRJPs is very likely to have an advantage effect on murine immune functions.
