Background Alcohol is a widely abused drug with many health implications, mainly caused by the oxidative and nitrosative stress on different body parts. Ayurvedic herbalism authenticates the multiple therapeutic applications of Terminalia arjuna bark due to its rich phytochemical repertoire. Objective To observe the extent of oxidative damage caused to erythrocytes by alcohol and assess the protective ability of T. arjuna bark powder aqueous extract (AETA) against the damage. Materials and methods Wister albino rats were categorized into four groups of eight rats per group; first group (control) was fed with glucose, second group was given alcohol at a dose of 20% v/v; 5g alcohol/kg b. wt/day, third group was co-administered with AETA (0.5 g/kg b. wt/day) and alcohol and the fourth group was kept on bark extract alone. Blood samples were collected and evaluated for different biochemical parameters after the completion of the treatment period. Results Alcohol significantly increased the erythrocyte membrane protein carbonyl and malondialdehyde (MDA) contents, along with a concomitant decrease in the membrane antioxidant status, when compared to the control group. Chromatographic analysis of the alcohol-treated rat erythrocyte membranes revealed altered membrane individual phospholipid contents and fluidity properties. Alcohol-induced morphological changes in the erythrocytes and its effect on decreasing the resistance of hypotonic shock induced by NaCl are evident from the hemolysis curves. However, AETA administration to alcoholic rats beneficially modulated the membrane properties anvd protected erythrocytes from damage. Conclusion Results suggest that AETA protects erythrocytes from alcohol-induced oxidative stress, biophysical, and biochemical changes very effectively.

Amaç: Kemoterapötik ilaçlar kanser hücrelerinin ortadan kaldırılmasında etkili iken aynı zamanda sağlıklı hücrelerde de hasar oluşturabilmektedir. Bu çalışmada, buğday çimi (Triticum aestivum L.) ekstraktının fenolik bileşen içeriğinin analizi ve bu ekstraktın kemoterapötik tedavide kullanılan sisplatin ve etoposid’in sağlıklı hücrelerde oluşturduğu DNA hasarına karşı etkisinin belirlenmesi amaçlanmıştır. Gereç ve Yöntemler: Çimlendirilmiş buğdayların metanol ekstraktı hazırlanarak HPLC (yüksek performanslı sıvı kromatografisi) ile fenolik bileşen analizi yapıldı. Buğday ekstraktı konsantrasyonuna bağlı hücre canlılık testi uygulanarak IC50 (Yarı maksimum inhibitör konsantrasyonu) ve LD50 (ortalama öldürücü doz) değerleri hesaplandı. Belirlenen bu konsantrasyon değerleri ile hücreler inkübe edilerek DNA hasarı varlığı Comet metodu ile değerlendirildi. Bulgular: Fenolik bileşen analizi sonucunda p-hidroksibenzoik asit en yüksek miktarda, o-kumarik asit ise en düşük düzeyde tespit edildi. Lenfosit hücrelerine uygulanan farklı konsantrasyonlardaki buğday çimi ekstraktı, etoposid ve sisplatin için değerler sırasıyla IC50=204,6 µg/mL, LD50=15,84 µg/mL ve LD50=24,51 µg/mL olarak bulundu. Comet analizi sonucunda kontrol grubuna kıyasla, etoposid LD50 ve etoposid LD50+buğday çimi ekstraktı IC50 grubu istatistiksel olarak anlamlı bulunurken (p>0,05), etoposid LD50 ve etoposid LD50+buğday çimi ekstraktı IC50 grubu arasında istatistiksel olarak anlamlılık bulunamadı (p>0,05). Bu sonuca benzer olarak kontrol grubuna kıyasla, sisplatin LD50 ve sisplatin LD50+buğday çimi ekstraktı IC50 grubu istatistiksel olarak anlamlı bulunurken (p>0,05), etoposid LD50 ve etoposid LD50+buğday çimi ekstraktı IC50 grubu arasında istatistiksel olarak anlamlılık saptanmadı (p>0,05). Sonuç: Çalışmamızda buğday çiminin etoposid ve sisplatin nedeni le oluşan DNA hasarında azalmaya neden olduğu görülmüş olmasına rağmen istatistiksel olarak anlamlılık saptanmamıştır.

Binge drinking (BD) is an especially pro‐oxidant pattern of alcohol consumption, particularly widespread in the adolescent population. In the kidneys, it affects the glomerular filtration rate (GFR), leading to high blood pressure. BD exposure also disrupts folic acid (FA) homeostasis and its antioxidant properties. The aim of this study is to test a FA supplementation as an effective therapy against the oxidative, nitrosative, and apoptotic damage as well as the renal function alteration occurred after BD in adolescence. Four groups of adolescent rats were used: control, BD (exposed to intraperitoneal alcohol), control FA‐supplemented group and BD FA‐supplemented group. Dietary FA content in control groups was 2 ppm, and 8 ppm in supplemented groups. BD provoked an oxidative imbalance in the kidneys by dysregulating antioxidant enzymes and increasing the enzyme NADPH oxidase 4 (NOX4), which led to an increase in caspase‐9. BD also altered the renal nitrosative status affecting the expression of the three nitric oxide (NO) synthase (NOS) isoforms, leading to a decrease in NO levels. Functionally, BD produced a hydric‐electrolytic imbalance, a low GFR and an increase in blood pressure. FA supplementation to BD adolescent rats improved the oxidative, nitrosative, and apoptotic balance, recovering the hydric‐electrolytic equilibrium and blood pressure. However, neither NO levels nor GFR were recovered, showing in this study for the first time that NO availability in the kidneys plays a crucial role in GFR regulation that the antioxidant effects of FA cannot repair.
