As an environmental contaminant, Benzo[a]pyrene (B[a]P; BaP) disrupts the antioxidant signaling and thus leads to the induction of oxidative stress and the damage of DNA in the ovary. low-dose atorvastatin (ATV) has antioxidant and anti-apoptotic properties. The present study aimed to survey the effects of prenatal exposure to BaP on ovarian toxicity and also to investigate the protective role of ATV in reducing ovarian toxicity. In this study, rats were divided into seven groups: control, ATV (10 mg/kg), oil, BaP (10 and 20 mg/kg), and ATV + BaP (10 and 20 mg/kg). BaP and ATV were administrated from gestation day 7-16 (GD7 to GD16), orally. 10 weeks after the birth, female offsprings were examined for oxidative stress markers, sex hormones, ovarian and tubular tissue structure, and the apoptosis markers. Data showed that BaP significantly reduced glutathione, increased malondialdehyde level, and disrupted the tissue structure of the ovary. Moreover, estrogen and progesterone levels significantly decreased in the offsprings rats. Also, BaP increased caspase-3 immunoreactivity. Atorvastatin treatment along with BaP in the embryonic period were able to bring the antioxidant status and sex hormones levels relatively close to normal. Besides, histological findings showed that atorvastatin was able to improve ovarian and oviduct abnormalities caused by BaP. Based on the above studies be concluded that atorvastatin in the embryonic during was able to reduce ovarian damage caused by BaP with antioxidant and anti-apoptotic properties.

Antioxidant and Nephroprotective Effects of Okra Pods Extract (Abelmoschus esculentus L.) against Lead Acetate-Induced Toxicity in Mice

In this study, we determine the curative effects of okra pods (Abelmoschus esculentus L.) extract against lead acetate toxicity in mice kidney. n-Hexane, ethyl acetate, and methanol solvent were used for extracting okra pods. The role of the extract as an antioxidant was tested by DPPH and FRAP methods. The methanol extract was used for experiments in animals. A total of 30 male BALB/c mice were randomly divided into six equal groups: normal control, negative control (lead-induced), and treatment groups (lead-induced for 28 days and administration of methanol extract at doses of 50, 100, 200, and 400 mg/kg BW for the 28 days). The following were analyzed in all groups: activity of the antioxidant enzymes, namely, superoxide dismutase (SOD) and catalase (CAT); oxidant level, namely, malondialdehyde (MDA) and nitric oxide (NO); and markers of kidney injury, namely, blood urea nitrogen (BUN) and creatinine (Cre). Kidney histopathology was also evaluated. This study showed that the methanol extract showed the highest antioxidant activity (IC50 is 35.21 µg/mL, and FRAP is 57.58 µM Fe2+/g). The CAT and SOD activities increased significantly in okra-treated groups (P<0.05). The okra administration groups experienced a significant decrease in MDA, NO, BUN, and Cre levels (P<0.05). Thickness of the epithelial proximal tubule, diameter of the proximal tubule, and percentage of necrotic cells in proximal tubule decreased, but the diameter ratio of glomerular Bowman’s capsule in mice treated with okra was optimally improved and repaired like normal control (P<0.05). The results of this study reveal that methanol extract has a very strong antioxidant effect and can reduce the influence of toxicity induced by lead acetate in mice kidney.

Objective: Cyclophosphamide (CP), as an anticancer agent has side effects, especially hepatotoxicity. Cerium oxide nanoparticles [nanoceria (NC)], which act as antioxidants, have a protective effect against oxidative stress-induced toxicity. This study evaluated the preventive effect of NC in hepatotoxicity induced by CP. Materials and Methods: Thirty-two mice were randomly distributed into four groups: control (received only normal saline), NC (received NC 100 μg/kg for 3 days, intraperitoneally), CP (received CP 200 mg/kg single dose on the third day of the study, intraperitoneally), and NC+CP (NC 100 μg/kg and CP 200 mg/kg). Two days after the final treatment, oxidative stress marker (MDA and GSH) and alanine aminotransferase (ALT), aspartate aminotransferase (AST), and alkaline phosphatase (ALP) levels were measured, and histopathological and immunohistochemical examinations were performed for determining the effects of NC on hepatotoxicity. Results: CP-induced hepatotoxicity was evident by the significant elevation of liver enzyme levels and alteration in oxidative stress marker levels. Additionally, histopathological changes and apoptosis were markedly increased. NC decreased MDA level, increased GSH level and decreased liver enzyme levels in CP-treated mice. In addition, NC pretreatment could be alleviated by the immunoreactivity of caspase-3 in CP-treated mice compared with the CP group. Conclusion: NC showed a potent antioxidant effect in hepatotoxicity induced by CP. The study suggested that NC through its antioxidant and anti-apoptotic properties has a protective effect against hepatotoxicity induced by CP.
