NETosis promotes cancer-associated arterial microthrombosis presenting as ischemic stroke with troponin elevation

Abstract: INTRODUCTION Large elevations of high sensitive Troponin T (hsTnT) in ischemic stroke patients is associated with a poor outcome. In a pilot study we found a high prevalence of malignancies among these patients. Since neutrophil extracellular traps (NETs) have been linked to cancer-associated thrombosis, we hypothesized that the concomitant cerebral and myocardial ischemia could be the result of a NET-induced hypercoagulable state. MATERIALS AND METHODS Clinical assessments, plasma analyses and autopsies wit… Show more

“…This is in line with a recent study revealing a low amount of H3Cit in plasma of healthy individuals when applying a similar assay but without concentrations derived from a standard curve [9]. Furthermore, in accordance with previous studies showing elevations of surrogate markers of NETs in septic patients [10, 11, 25, 26] as well as the detection of H3Cit by western blotting in murine models of LPS-induced septic shock [27, 28], our results support an inflammatory induction of a systemic NET burden by showing clear intra-individual elevations of H3Cit in healthy individuals receiving LPS injection.…”

“…In summary, we suggest that neutrophils should continue to be explored as therapeutic targets in APS. There is also a need to extend these studies to models beyond venous thrombosis, including arterial thrombosis and thrombosis in the microvasculature (10, 15, 48). …”

“…This is less common than simply including diagnoses prior to stroke, but the definition varies widely in existing literature [8, 13, 26, 33, 34]. However, based on the results from our initial analyses showing no significant differences among patients diagnosed with cancer pre- and post-stroke, both groups were included as patients with “active cancer”.…”
