Network pharmacological identification of active compounds and potential actions of Erxian decoction in alleviating menopause-related symptoms

Abstract: BackgroundErxian decoction (EXD) is used to treat menopause-related symptoms in Chinese medicine. This study aims to identify the bioactive compounds and potential actions of EXD by network pharmacological analysis.MethodsTwo databases, the Traditional Chinese Medicine Systems Pharmacology database and TCM Database@Taiwan, were used to retrieve literature of phytochemicals of EXD. STITCH 4.0 and the Comparative Toxicogenomics Database were used to search for compound–protein and compound–gene interactions, res… Show more

“…For several compounds that are not included in TCMSP database but are found in TCM Database@Taiwan ( ), Lipinski’s Rule (LR) was used for active compound identification according to the following criteria: molecular weight (MW) ≤ 500, chemical composition with not more than ten hydrogen bond acceptors (Hacc ≤ 10), lesser than five hydrogen bond donors (Hdon ≤ 5), and octanol-water partition coefficient lesser than five (LogP ≤ 5) [ 22 , 27 ]. Compounds that did not satisfy at least two of the above requirements were excluded.…”

“…The human protein–protein interaction (PPI) network analyzed here included 15,554 proteins and 642,304 PPIs, which was constructed using the data provided in STRING (Szklarczyk et al, 2015 ). In order to ensure the reliability of the analyzed data, only those PPIs with high confidence score (>0.95) were collected for the subsequent analyses (Ghosh et al, 2015 ; Wang S. et al, 2015 ). As a result, a sub-network with 8,509 proteins and 40,468 PPIs were generated and adopted for further analyses in this study.…”

“…TCM decoctions have complex ingredients and multiple targets, and network pharmacology can predict novel compound targets and the potential pathways of action based on existing TCM decoctions and has helped clarify the mechanism of several TCM decoctions so far [21][22][23].…”
