Spontaneous abortion (Sa) occurs in woman of child-bearing age, jeopardizing their physical and mental health. Quercetin is a natural flavonoid, which exhibits a variety of pharmacological activities. However, the role and mechanisms of quercetin in Sa still need to be further explored. animal experiments were performed to examine the effect of quercetin in treating Sa. institute of cancer research mice were injected with lipopolysaccharide into the tail vein on the 7th day of gestation to establish a Sa model. Gavage was performed during days 3-8 of gestation with high-, medium-and low-dose of quercetin. Then the effect of quercetin on embryos was evaluated. animal experiment showed that quercetin could remarkably reduce the embryo loss rate and increase the mean weight of surviving embryos to some degree. Furthermore, network pharmacology was employed to explore the underlying mechanisms of quercetin in the treatment of Sa. Several databases were used to collect the targets of Sa and quercetin. Protein-protein interaction network, Gene ontology and Kyoto encyclopedia of Genes and Genomes pathway enrichment analysis were performed to elucidate the interactions between Sa and quercetin. The relative mrna expressions of several targets in uterine were detected by quantitative reverse transcriptase polymerase chain reaction (rT-qPcr). network pharmacology indicated that the effects of quercetin in treating Sa were mainly related to hormone response and the modulation of defense response and inflammatory response, involving signaling pathways such as Pi3K-akt, VeGF, MaPK and core targets such as aKT1, albumin, caspase-3. rT-qPcr showed that quercetin could up-regulate aKT1, MaPK1, PGr, SGK1 and down-regulate eSr1, MaPK3. The results showed that quercetin may modulate multiple signaling pathways by targeting core targets to prevent and treat Sa.

BACKGROUND: Gastric mucosal injury caused by ethanol is a common gastrointestinal disease. Quinoa (Chenopodium quinoa Willd.), as a nutrient-rich grain, plays a significant role in preventing and treating gastric mucosal damage. The present study aimed to explore the protective effect of quinoa on alcohol-induced gastric mucosal damage and its possible mechanism.RESULTS: The ethanol-induced gastric mucosal injury rat model was used for in vivo experiments and H 2 O 2 -induced GES-1 cells for in vitro experiments to elucidate the protective effect of quinoa. The results show that quinoa water extract can increase the superoxide dismutase level and decrease the malondialdehyde level in vitro and in vivo. Furthermore, quinoa also reduced the bleeding point and bleeding area in rats with ethanol-induced gastric mucosal injury and improved gastric histopathological changes. H 2 O 2 significantly increased the levels of inflammatory factors in GES-1 cells, which were markedly ameliorated by quinoa water extract. Likewise, quinoa water extract regulated the protein expression levels of Nrf2, Keap1, HO-1, p-IKK, and p-NF-κB through Nrf2 and nuclear factor-κB signaling pathways, reducing the production of oxidative stress and inflammation, thereby repairing the damaged gastric mucosa. CONCLUSION:The findings of this study demonstrated that quinoa shows protective effect against ethanol-induced gastric mucosal injury through its anti-inflammatory and anti-oxidant effects. We propose that our research will provide a reference for quinoa as a functional food.

Osteoporosis is a serious threat to human life. Guben Zenggu Granule is an empirical prescription for clinical treatment of osteoporosis. MC3T3-E1 cells are mouse osteogenic precursor cells with osteogenic differentiation, and are classic cells for studying bone metabolism and osteogenic mechanism, as well as mechanical stimulation sensitive cells. Therefore, it can be inferred that Guben Zenggu granule can repair MC3T3-E1 cells under continuous static pressure overload. This study aims to through the network of pharmacology and gene sequencing method, reveal thrift increase bone particles under the condition of continuous static pressure overload on osteogenesis mechanism of MC3T3-E1 cells. In the process of analysis, from a variety of 98 compounds was predicted in the database, a collection of 474 goals, a total of 29,164 difference between two groups of genes. Then, construction of composite targets between cells and predict targets and protein - protein interaction networks, and through the cluster analysis to further explore the relationship between the target. In addition, linkages between target proteins and cells were further identified using Gene Ontology (GO) and Pathways (KEGG Pathway). Finally, the repair effect of Guben Zenggu granule on MC3T3-E1 cells under continuous static pressure overload was verified through experiments, so as to accurately explain the pharmacodynamic mechanism of Traditional Chinese medicine.
