Neuromuscular blocking activity of a new series of quaternary N‐substituted choline esters

Abstract: Summary1. The neuromuscular blocking activity of a new group of mono-and diquaternary N-substituted choline esters of several carboxylic acids has been evaluated in various whole animal preparations in the mouse, rat and cat. 2. A basic non-depolarizing mechanism was present in all but one compound -a depolarizing agent, which was studied for comparison. There was evidence of facilitatory activity and nicotinic stimulation in the monoquaternary compounds. These effects were diminished in the diquaternary compo… Show more

“…The highest toxicity was observed at iv injection (LD 50 0.51 mg/kg). The LD 50 of rocuronium at iv injection to rats is about 0.3 mg/kg [ 30 ], for (+)-tubocurarine in mice—0.11 mg/kg [ 31 ] and for vecuronium in mice—50 µg/kg [ 32 ]. In our experiment, intramuscular injection of rocuronium at dose of 0.13 mg/kg resulted in the death of the animal, while Az at 0.7 mg/kg was not lethal.…”

“…Since neuromuscular blockade has a quantal response to drug concentration the pharmacodynamics are suitably described by a logprobit dose-response relationship (Ginsburg, Kitz and Savarese, 1971). Construction of cumulative dose-response curves showed Org NC45 to be about 15% more potent than pancuronium and about five times more potent than atracurium for doses producing 95% depression of twitch response.…”

“…He developed the “train‐of‐four” (TOF) stimulation method to quantify the degree of neuromuscular blockade, and built a prototype for a clinical “TOF” neuromuscular monitor in Liverpool . Dr. Savarese also popularized the log‐probit plot of the dose response of neuromuscular blockers, a technique which converted an arithmetic sigmoid dose‐response curve into a straight line. Applying these methods of measurement and analysis allowed for accurate comparison of the potency and modes of action of different drugs.…”
