Neuroprotective Actions of Different Exogenous Nucleotides in H2O2-Induced Cell Death in PC-12 Cells

Abstract: Exogenous nucleotides (NTs) are considered conditionally essential nutrients, and the brain cannot synthesize NTs de novo. Therefore, the external supplementation of exogenous NTs is of great significance for maintaining normal neuronal metabolism and function under certain conditions, such as brain aging. This study, therefore, sets out to assess the neuroprotective effect of four kinds of single exogenous NTs and a mixture of the NTs, and to elucidate the potential mechanism. A rat pheochromocytoma cell line… Show more

“…In particular, indomethacin causes gastric injury by decreasing the content of antioxidants (e.g., SOD and GSH) and increasing lipid peroxidation products (e.g., MDA) [ 38 ]. SOD and GSH are important enzymes that scavenge superoxide anion radicals and protect cells from oxidative damage [ 39 ]. MDA cross-links DNA and proteins, disturbs cell division, and destroys the structure and function of proteins and enzymes [ 40 ].…”

“…Intestinal mucosa lacks the ability to produce NTs [22], so supplementation with NTs is important for intestinal health. Exogenous NTs can reduce the level of intracellular inflammatory phenotypes [23], and can also suppress inflammation in vivo [24,25].…”

“…Superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) are critical components of the endogenous antioxidant defense mechanism, which have the function of scavenging free radicals, chelating metal ions, and mitigating oxidative damage [ 28 , 29 , 30 ]. Malondialdehyde (MDA) is the key factor that causes damage to the biofilm system during lipid peroxidation and is an important indicator for evaluating oxidative stress damage [ 29 , 30 , 31 ]. The experimental results show that the POP intervention significantly increases the activity of SOD in the myocardium and gastrocnemius muscle and decreases the level of myocardial MDA available, indicating that the POPs can exert anti-fatigue effects by improving the antioxidant capacity and reducing oxidative stress damage in the body.…”
