Neuroprotective and Anti-Inflammatory Effects of the Flavonoid-Enriched Fraction AF4 in a Mouse Model of Hypoxic-Ischemic Brain Injury

Abstract: We report here neuroprotective and anti-inflammatory effects of a flavonoid-enriched fraction isolated from the peel of Northern Spy apples (AF4) in a mouse of model of hypoxic-ischemic (HI) brain damage. Oral administration of AF4 (50 mg/kg, once daily for 3 days) prior to 50 min of HI completely prevented motor performance deficits assessed 14 days later that were associated with marked reductions in neuronal cell loss in the dorsal hippocampus and striatum. Pre-treatment with AF4 (5, 10, 25 or 50 mg/kg, p.o… Show more

“…The peels of the apple cultivar Northern Spy were collected from a commercial pie manufacturer, Apple Valley Foods Inc. (Kentville, NS, Canada) and isolation of AF4 was performed as previously described in Keddy et al (24). Briefly, after peeling, the peels were treated with 2% CaCl 2 in water (w/v) at 55 § 5 C for 10 min to prevent degradation of phenolic compounds.…”

“…Eluates were concentrated to 20 ml using a rotary evaporator (Rotavapor Ò R-200; Buchi, Flawil, Switzerland) at 45 C. AF4 had the highest proportion of flavonols and other monomeric phenolic compounds and was therefore selected for this study. LC-MS/MS analysis was performed to analyze monomeric polyphenols present in AF4 (Table 1) according to our previously described procedure in Keddy et al (24).…”

“…After incubation, the sections were rinsed in water and destained with 1 % acetic acid solution and finally dehydrated by rinsing with ethanol. The slides were rinsed thoroughly and observed under microscopy, photographed and analysed [16].…”

“…Apple peel is a particularly rich source of flavonoids and contain 3-6 fold more flavonoids than the flesh 9 . Apple peel extracts are known to act as antioxidants 10 , antiviral 11 , neuroprotective 12 , antiproliferative and anticancer agents 4,13 . Since apple peel is a waste product of apple processing industry, there is potential to convert this bioresource into a natural health product to treat hypertension 14 .…”
