Neuroprotective Effects of Ginsenoside Rg3against 24-OH-cholesterol-induced Cytotoxicity in Cortical Neurons

Abstract: Ginsenoside Rg 3 (Rg 3 ), one of the active ingredients in Panax ginseng, attenuates NMDA receptor-mediated currents in vitro and antagonizes NMDA receptors through a glycine modulatory site in rat cultured hippocampal neurons. In the present study, we examined the neuroprotective effects of Rg 3 on 24-hydroxycholesterol (24-OH-chol)-induced cytotoxicity in vitro. The results showed that Rg 3 treatment significantly and dose-dependently inhibited 24-OH-chol-induced cell death in rat cultured cortical neurons, … Show more

“…The cytotoxic effect of lancemaside A was then evaluated by a conventional MTT assay as reported previously [28, 29]. For the final 3 h of culture, 10  μ L MTT solution (10 mg/mL in phosphate buffered saline, pH 7.4) was added to each well.…”

“…Three hours prior to culture termination, 10 ml of a MTT solution (10 mg/ml in phosphate buffered saline, pH 7.4; PBS) was added and the cells were continuously cultured until termination of the experiment. Incubation was halted by the addition of 15% sodium dodecyl sulphate into each well to solubilize the produced formazan (Roh et al, 2010). The absorbance at 570 nm (OD 570 ) was measured using a Spectramax 250 microplate reader.…”
