Neutral sphingomyelinase‐2 and cardiometabolic diseases

Abstract: Sphingolipids, in particular ceramides, play vital role in pathophysiological processes linked to metabolic syndrome, with implications in the development of insulin resistance, pancreatic ß-cell dysfunction, type 2 diabetes, atherosclerosis, inflammation, nonalcoholic steatohepatitis, and cancer. Ceramides are produced by the hydrolysis of sphingomyelin, catalyzed by different sphingomyelinases, including neutral sphingomyelinase 2 (nSMase2), whose dysregulation appears to underlie many of the inflammation-re… Show more

“…SM is primarily produced in the Golgi apparatus and transferred to all other cellular membranes ( Campelo et al, 2017 ). Sphingomyelinase catalyzes the hydrolysis of the phosphodiester bond of sphingomyelin and yields Cer and PC after stimulation by TNF-α or IL-1β ( Sindhu et al, 2021 ). In our sample, SM disorders dominated the lipid abnormalities observed in the MP subgroup and thus provide possible treatment targets for MP.…”

“…Neural sphingomyelinase 2 (nSMase2) is a hydrolase enzyme that involved in sphingolipid metabolism reactions ( Sindhu et al, 2021 ). Nobuyoshi Kosaka et al demonstrated that inhibiting the activity of nSMase2 lead to reduced exosomal miRNA secretion.…”

“…The phosphorylation of sphingosineby-sphingosine kinases (SphK1 and SphK2) produces sphingosine 1-phosphaet (S1P), a molecule involved in angiogenesis, proliferation, migration, transformation, and inflammation of cells. This SL turnover pathway has been estimated to contribute from 50% up to 90% of the ceramide biosynthesis [31,32]. The salvage pathway is not only subject to regulation, but it also modulates the formation of ceramide and subsequent ceramide-dependent cellular signals [33].…”
