New Antimycobacterial Saponin from Colubrina retusa

Abstract: A new jujubogenin saponin was isolated from the stems of Colubrina retusa and identified as jujubogenin 3-O-alpha-L-arabinofuranosyl (1-->2)-[3-O-(trans)-p-coumaroyl-beta-D-glucopyranosyl (1-->3)]-alpha-L-arabinopyranoside (4) on the basis of chemical and spectroscopic data. The antimycobacterial activity expressed as minimum inhibitory concentration (MIC) for compound 4 was 10 microg/mL.

“…
Stevioside and steviolbioside, glycosides from Stevia rebaudiana Bertoni, in addition to hybrid compounds synthesized from steviolbioside and the antituberculosis drug isoniazid and its isomer nicotinic acid hydrazide exhibited moderate antituberculosis activity against M. tuberculosis strain H 37 R V in vitro (MIC = 7.5, 3.75,5, and 10 Pg/mL, respectively).

“…Solsodomine A, a pyrrole alkaloid isolated from Solanum sodomaeum exhibited antimycobacterial activity against M. intracellulare with 10 µg/mL MIC value [27]. Two flavonoids linaroside and lantanoside were isolated from Lantana camara exhibited antimycobacterial activity against M. [31]. From aerial part of Ruprechtia triflora, 5α,8α-epidioxyergosta-6,22-dien-3β-yl stearate which a novel sterol was isolated and exhibited antimycobacterial activity with the MICs of 2-128 µg/mL [32].…”

“…A new saponin jujubogenin, isolated from Colubrina retusa L. (Rhamnaceae), had antimycobacterial activity at minimum inhibitory concentration (MIC) of 10 μg/ml when tested against Mycobacterium intracellulare. 86 It has been documented that triterpenoid saponins from other sources, such as Maesalanceolata, Maesachisia, and Maesaindica, display direct virucidal activity against Newcastle disease virus, vaccinia virus, and herpes simplex virus. 87 Several saponins, for example aescine from Aesculus hippocastanum, primula saponin from Primula veris, saikosaponin A from Bupleurum falcatum, theasaponin from Thea sinensis, and gymnemic acid from Gymnema sylvestre, showed antagonist activity against influenza A2 virus.…”
