New classification of rheumatoid arthritis based on immune cells and clinical characteristics

Abstract: Background Rheumatoid arthritis (RA) is a chronic systemic immune disease characterized by joint synovitis, but the specific etiology is unknown, and the characteristic serum diagnostic markers are also lacking. Methods First, we obtained the gene expression profile of synovium to evaluate the infiltration of immune cells in synovium, and screened the differentially expressed immune related genes for enrichment analysis. Subsequently, we classified RA into three subtypes by unsupervised clustering of serum gen… Show more

“…These conditions have different HLA-based risk alleles, disease mechanisms, and disease presentations compared to seropositive conditions (46). While these seronegative conditions are not classically associated with a positive ANA, individuals with these conditions can have higher rates of ANA positivity compared to the general population (47)(48)(49) and often have an ANA test ordered as part of their clinical evaluation (26). In a sensitivity analysis, not counting the individuals with seronegative conditions as cases did not greatly impact the performance of the model.…”

“…In addition to causing joint swelling, pain, stiffness and deformities, it can also damage multiple systems of the body, including the skin, lungs, heart and blood vessels, causing great harm to patients 4 . The exact cause of RA is not yet clear, and factors such as immunity, infection and genetics are closely related 5,6 …”

“…4 The exact cause of RA is not yet clear, and factors such as immunity, infection and genetics are closely related. 5,6 The stimulation of abnormal chronic inflammation caused by RA, genetic susceptibility and regulation of immune response in RA treatment may all be related to the occurrence of malignant tumours. 7 Mechanistically speaking, both diseases exhibit abnormal immune activation and inflammatory processes.…”
