New Method for Simultaneous Determination of Microcystins and Cylindrospermopsin in Vegetable Matrices by SPE-UPLC-MS/MS

Abstract: Cyanotoxins are a large group of noxious metabolites with different chemical structure and mechanisms of action, with a worldwide distribution, producing effects in animals, humans, and crop plants. When cyanotoxin-contaminated waters are used for the irrigation of edible vegetables, humans can be in contact with these toxins through the food chain. In this work, a method for the simultaneous detection of Microcystin-LR (MC-LR), Microcystin-RR (MC-RR), Microcystin-YR (MC-YR), and Cylindrospermopsin (CYN) in le… Show more

“…UPLC-MS/MS-based methods provide the possibility of simultaneous detection of different congeners in various sample types [20,21,22,23,24,25]. However, quantitation from complex matrices like blood or tissue homogenate is difficult due to matrix residues which may influence the signal (suppression or enhancement), thus leading to under- or over-estimation of the true amount of MC present.…”

“…As a first step, a C 18 reversed-phase HPLC column (LichtoCART ® ) was employed to optimize the separation conditions of the 10 selected cyanotoxins. MeOH and ACN acidified with FA have been used in general as organic phases in most of the analytical methods previously reported [29] (especially ACN). In Table S2 of the Supplementary Materials, several chromatographic parameters were compared.…”

“…The transitions employed for setting up the UPLC-MS/MS system were: 996.5/135.0, 996.5/213.1, and 996.5/996.5 for MC-LR; 416.2/194.0 and 416.2/176.0 for CYN. The first values for each toxin were used for quantification, while the others for confirmation [66].…”
