New polymorphic microsatellite markers in the human MHC class II region

Abstract: The human major histocompatibility complex (MHC) class II region spans approximately 1.1 Mb and presently contains over 30 functional genes Susceptibility loci to numerous diseases, mainly of autoimmune nature are known to map to the this region, as assessed by associations with particular HLA class II alleles. However, it has been difficult to precisely localize these susceptibility loci to a single gene, for example DQB1 or DRB1, due to the tight linkage disequilibrium observed in the HLA class II region. To… Show more

“…The RPD [55][56][57] epitope is associated with protection; however, the individual residues R, P, and D, when evaluated independently, are not. This is reminiscent of the association of rheumatoid arthritis with the residues DR␤ 67 , DR␤ 70 , DR␤ 71 , and DR␤ 74 together, but not any of these residues independently (42).…”

“…Regardless of the role of this DR epitope, whether it is part of a binding pocket of an HLA molecule or part of a peptide bound to an HLA molecule, this epitope operates as a defined sequence and not as individual residues. The RPD [55][56][57] epitope of this study is also located in a position that is known to influence peptide binding and T cell recognition. Residue 57 participates in the formation of a pocket 9 of DQ molecules and is associated with susceptibility to insulindependent diabetes mellitus.…”

“…More than 1000 polymorphic microsatellite markers have been described within the HLA region. [13][14][15][16] The microsatellite markers showed considerable polymorphism and strong LD with particular alleles of classical HLA loci composing of well-defined extended HLA haplotypes. 17 The HLA haplotypes can be separated into several blocks, including a haplotype block containing the HLA-Cw and -B genes, just centromeric to the MHC class I region, which is known to be one of the highest polymorphic loci in the human genome.…”
