New Polyoxygenated Briarane Diterpenoids, Briaexcavatolides O−R, from the Gorgonian Briareum excavatum

Abstract: Four new polyoxygenated briarane-type diterpenoids, briaexcavatolides O-R (1-4), have been isolated from a gorgonian octocoral Briareum excavatum. Their structures were determined using spectroscopic and chemical methods. Metabolites 1-3 were found to contain oxygenated substituents at C-2, C-3, and C-4, and the relative configurations were assigned as 2R*,3R*,4R* at these three positions. Briaranes containing this type of stereochemistry are reported for the first time. The structures of metabolites 1 and 2 w… Show more

“…The molecular formula of C 28 Tables 1, 2) and 2D-NMR (Fig. 7, Table 3) data of 4 were similar to those of a known briarane derivative, briaexcavatolide P (7), 33) except that the signals corresponding to a butyryloxy group in 7 were replaced by signals for an acetoxy group in 4. The correlations from a NOESY experiment of 4 ( Fig.…”

“…Thus, the remaining hydroxy groups had to be positioned at C-12 and C-14 by the key 1 H-1 H COSY correlations and characteristic NMR signal analysis. The relative stereochemistry of 3 was confirmed as being similar to that of a known metabolite, briaexcavatolide P (7), 33) by comparison of the NMR chemical shifts and coupling constant analysis for the chiral centers. In the NOESY experiment of 3 (Fig.…”

“…Five briarane analogues, brianthein W (627), 9-deacetylbriareolide H (628), 2β-acetoxy-2-(debutyryloxy) stecholide E (629), 9-deacetylstylatulide lactone (630), and 4β-acetoxy-9-deacetylstylatulide lactone (631) [274], were isolated from an unidentified species of the genus Briareum which is known for generating the first briarane diterpenoid, briarein A [275]. Extensive chemical investigation on B. excavatum resulted in the isolation of a large group of briaranes, namely the excavatolides A-M (632-644) [276,277] and U-Z (645-650) [278], the briaexcavatolides A-Z (651-676) [279][280][281][282][283][284], as well as the brianolides 677 and 678 [276,280]. The natural briarane diterpenoids are commonly cytotoxic compounds.…”

In continuation of our interest in the chemistry and potential medicinal importance of briaranes, [12][13][14] we investigated the gorgonian octocoral Junceella juncea collected in Taiwan.…”
