New record species of Puntius (Pisces: Cyprinidae) from West Sumatra based on Cytochrome Oxidase1Gene

Abstract: Biodiversity study on Puntius has been conducted in West Sumatera using a molecular technique. From the genetic analysis using the sequence of CO1 gene, the study discovered: (1) A new record species of Puntius in Diatas Lake, Batang Lembang, Batang Gumanti, Muara Pingai rivers (located in the eastern part of the Bukit Barisan mountain range) which is Barbodes binotatus banksi or B. banksi. (2). A new record of subspecies in Maninjau Lake and its tributary (located in the western part of the Bukit Barisan moun… Show more

“…The average of nucleotide composition was of A: 27.30%, C: 27.20%, T: 29.40%, and G: 16.10%. The nucleotide composition was similar to previous studies (Negi et al 2017;Roesma et al 2018). The frequency of distributions for the four nucleotide bases was significantly different, with nonoverlapping confidence intervals.…”

“…All populations of P. cf. binotatus in the first subcluster are located at the highland over 1.500 meters above sea level in the eastern part of Bukit Barisan Mountain range (Roesma et al 2018). Bukit Barisan Mountain range is the row of the mountain that spreading out from northern to southern of Sumatra island more or less 1650 km in length.…”

“…Geographically, the three lakes are not connected, no gene flow between the population of the lakes, that causes the geographical isolation. Roesma et al (2018) explained that the freshwater fauna in Maninjau Lake isolated because of no connection with other waters. Water in the Maninjau Lake comes from small rivers, which is filled by water catchment in the surrounding area and only has one outlet.…”

“…Mitochondrial DNA (mtDNA) markers have been widely used for most systematic molecular studies because they have a large number of copies, maternally inherited, and have a higher mutation rate compared to nuclear DNA (Brown et al 1979;Johns and Avise 1998). Mitochondrial cytochrome oxidase subunit I (COI) gene could serve as a rapid and reliable barcoding marker for identifying species and effective in delimitating species of various animals (Hebert et al 2004;Hubert et al 2008;Imtiaz et al 2017;Roesma et al 2018;Roesma et al 2019). Recently, the COI gene has been widely used to resolve the taxonomy in a gobioid group (Jeon et al 2012;Jin et al 2014;Taillebois et al 2014;Wang et al 2017;Olii et al 2019).…”
